摘  要:目的探讨小鼠肺爆震伤后不同时间点超氧化物歧化酶(SOD)-1、丙二醛(MDA)-5及肌醇酶(IRE)-α表达的变化。方法将64只小鼠分为空白对照组(8只)与爆震伤实验组(56只)。爆震伤模型建立后,分别于3、6、12、24、48 h与1、2周7个时间点解剖8只小鼠。观察每个时间点小鼠的呼吸与被毛状态等,应用RT-PCR与Western-blot检测小鼠肺组织SOD-1、MDA-5及IRE-α在基因与蛋白水平上的表达量。结果爆震伤后,小鼠肺组织SOD-1表达在3 h后开始降低,1周后上升;MDA-5表达在3 h后开始上升,12 h时达到最高,24 h后下降,1周后再次上升;IRE-α表达在3 h后开始上升,24 h时达到最高,48 h后下降,2周时恢复正常。结论小鼠肺组织在发生爆震伤3 h后开始出现氧化应激反应,12~48 h时氧化应激反应达到高峰,随后下降,到2周时基本恢复正常。Objective To investigate the changes of superoxide dismutase ( SOD ) -1, malonaldehyde (MDA) -5 and inositol enzyme (IRE) -α expression in rats at different time points after injury. Methods A total of 64 cases of rats were divided into the blank control group(n = 8) and exploratory shock group( n = 56). The model was established at 3 hours,6 hours, 12 hours,24 hours,48 hours,1 week and 2 weeks on 8 rats. The respiratory and hair status of rats at each time point were observed. The expression of SOD-l, MDA-5 and IRE-α in lung tissue were detected by RT-PCR and Western-blot. Results The expression of SOD-1 began to reduce after 3 hours of blast injury, and increased after 1 week. The expression of MDA-5 increased after 3 hours,reached the peak after 12 hours and reduced after 24 hours,then once again increased af- ter 1 week. The expression of IRE-α increased after 3 hours, reached the peak after 24 hours, reduced after 48 hours and re- turned to normal after 2 weeks. Conclusion Oxidative stress begins to occur after 3 hours of lung injury, and reaches the peak at 12 -48 h,then reduces and returns to normal at 2 weeks.
