摘 要:目的分析颅脑爆震伤(bTBI)后,不同时间点小鼠脑组织中超氧化物歧化酶(SOD-1)、一氧化氮合酶(NOS1)和内质网跨膜蛋白肌醇酶1α(IRE1α)的表达变化,探讨相关因子与时间的关系。方法健康昆明小鼠63只随机分为模型组和正常对照组。按照伤后时间将模型组分为3 h组、6 h组、12 h组、24 h组、48 h组、7 d组和14 d组。采用Evans blue染色、Western-blot和Real-time PCR方法,观察大脑损伤及测定伤后各组小鼠脑组织中SOD-1、NOS1和IRE1α的含量及m RNA的表达变化。结果与正常对照组相比,模型组小鼠脑组织Evans blue染色呈明显蓝色。与对照组相比,模型组小鼠脑组织各个时间点氧化应激指标SOD-1、NOS1和IRE1α含量的表达均显著增高,48 h后表达量有所降低,但仍高于正常组,差异有统计学意义(P<0.05)。结论 SOD-1、NOS1和IRE1α等指标参与脑爆震伤的发生和发展,监测相关指标变化对阐明b TBI致脑损伤的发生机制具有一定的意义。Objective To investigate the changes of expression of SOD-1, NOS1 and IRE1 α in mouse brain at different time points of blast-related traumatic brain injury( bTBI), and investigate the relationship between correlation factors and time. Methods A total of 63 healthy male rats were randomlydivided into the model group and the normal control group. Accord- ing to the injury time,the rats in the model group were divided into the groups of 3 hours,6 hours, 12 hours,24 hours, 48 hours,7 days and 14 days. Evans blue,Western-blot and Real-time PCR methods were used for determination injury and expression changes of SOD-1, NOS1 and IRElα inbrain tissue. Results Compared with normal control group, the Evans blue staining of brain tissue in model group was in significantly blue. The expression of SOD-1, NOS1 and IRElα at different time points in brain tissue of model group were significantly increased and reduced after 48 hours, while still higher com- pared to the normal control group(P 〈 0. 05). Conclusion SOD-1, NOS1 and IRE1 α involves in the occurrence and devel- opment of brain blast injury ,which has certain significance to monitor the changes of related indicators clarifying the mecha- nism of bTBI induced brain iniury.
