摘 要:为研究牛冠状病毒(Bovine Coronavirus,BCV)HE基因进化关系,试验利用提取牛冠状病毒核酸,将RNA反转录成cDNA,并以此为模板扩增出全长为1 300 bp HE基因片段,将其与pMD-18T载体连接,产物克隆入DH5α感受态细胞,测序,结果与Gen Bank中已发表的14株BCV的HE基因序列进行系统进化树分析。扩增出的牛冠状病毒HE基因序列(HLJ-14),符合HE基因型参照序列特点,与Gen Bank中已发表的另外14株BCV HE基因型全序列同源性为97.3%~98.8%。The bovine coronavirus nucleic acid was extracted to study the bovine coronavirus HE gene evolution. The RNA was transcribed reversely into cDNA which was amplified as a template to a length of 1 300 bp HE gene fragment ,connected with pMD-18T vector,cloned into DH5ot.It was sequenced and analysed the evolution tree with 14 strains BCV HE gene of the sequences published in GenBank. Amplification of bovine coronavirus HE gene sequence (HLJ-14) ,according with the characteristics of the HE genotype reference sequence,and GenBank had been published in the other 14 strains of BCV HE gene sequence homology of 97.3%-98.8%.
