滋阴清热方对系统性红斑狼疮小鼠细胞焦亡相关因子的影响  

The Effect of Ziyin Qingre Formula (滋阴清热方) on Pyroptosis Related Factors in Mice with Systemic Lupus Erythematosus

摘　　要：目的探讨滋阴清热方治疗系统性红斑狼疮的可能作用机制。方法 30只狼疮鼠随机分为中药组、脂多糖组和模型组各10只,另以正常C57BL/6J健康小鼠10只为空白组。中药组给予滋阴清热方20 g/(kg·d)灌胃,每日1次;脂多糖组给予脂多糖8 mg/kg腹腔注射,2天1次;模型组和正常组给予等容积灭菌蒸馏水灌胃,每日1次,各组均干预6周。检测各组小鼠肾组织半胱天冬氨酸蛋白酶1(Caspase-1)、白细胞介素1β(IL-1β)蛋白及核苷酸结合寡聚化结构域样受体蛋白3(NLRP3)、Caspase-1和IL-1βmRNA表达。结果与空白组比较,模型组小鼠肾组织Caspase-1、IL-1β蛋白表达升高,NLRP3、Caspase-1和IL-1βmRNA表达亦升高(P<0.05)。与模型组比较,中药组小鼠肾组织IL-1β蛋白及mRNA表达降低,而脂多糖组均升高(P<0.05);中药组Caspase-1和NLRP3 mRNA表达与模型组比较差异无统计学意义(P>0.05)。结论滋阴清热方可降低狼疮鼠细胞焦亡炎症因子,且可能是其治疗系统性红斑狼疮的作用机制之一。Objective To explore the possible action mechanism of Ziyin Qingre Formula（ 滋阴清热方） in treating systemic lupus erythematosus. Methods Thirty lupus mice were randomly divided into a Chinese medicine group,a lipopolysaccharide group and a model group,with 10 mice in each. Other 10 normal C57BL/6JN healthy mice were taken as the blank group. The Chinese medicine group was given 20 g/（ kg·d） of Ziyin Qingre Formula for gavage,once per day; the lipopolysaccharide group was given 8 mg/kg of lipopolysaccharide for intraperitoneal injection,once every two days; and the model and the normal group were given equivalent sterilized distilled water for gavage,once every day. Each group was treated for 6 weeks. Expressions of protein of cysteine containing aspartate 1（ Caspase-1） and interleukin 1β（ IL-1β）,mRNA of nucleotide binding oligomerization domain like receptor 3（ NLRP3）,Caspase-1 and IL-1β in kidney tissues of mice in each group were examined. Results Compared with those in the blank group,expression of protein of Caspase-1 and IL-1β,as well as mRNA of NLRP3,Caspase-1 and IL-1βin kidney tissues of mice in the model group increased（ P 0. 05）. Compared with that in the model group,expression of IL-1β protein and mRNA decreased in the Chinese medicine group,but increased in the lipopolysaccharide group（ P 0. 05）. There was no significant difference in Caspase-1 and NLRP3 mRNA expression between the Chinese medicine group and the model group（ P 0. 05）. Conclusion Ziyin Qingre Formula could decrease pyroptosis and inflammatory factors,which may be one of its action mechanisms in treating systemic lupus erythematosus.
