摘 要:以8个新疆芜菁(Brassica rapa L.)品种为试材,对影响小孢子培养中胚胎发育的影响因素进行了研究。结果表明,不同基因型间小孢子胚胎发生能力差异很大,在接种的8个基因型中,有4个诱导出胚,诱导成功率50%,其中W4出胚率最高,达到73胚/30蕾。NLN-13培养基中添加激素能明显促进胚状体的发生;低温预处理也能提高小孢子产胚率,最适宜的时间为24 h。In order to optimize microspore culture techniques of Brassica rapa L. and improve the embryo induction ra te, the influence factors to microspore embryogenesis of Brassica rapa L. were studied through eight genotypes. Results showed that there was significant difference in embryogenesis ability among different genotypes. Among them, four genotypes yielded em-bryos successfully, which the rate reached 50%. Genotype W4 was an ideal material for embryoid induction, and the fre-quency reached 73 embryos per 30 buds. NAdding phytohormone 6-BA and NAA in NLN-13 medium can improve the em-bryoid induction,4 ℃ pretreatment of bud for 24 h can significantly improve the production rate of microspore embryogenesis.
