摘 要:目的探索传统中药复方四君子汤与临床常用化疗药物氟尿嘧啶(5-FU)联用对体外培养的结肠癌HT-29细胞的生长抑制作用,为临床上应用四君子汤作为5-FU的增效剂联合治疗结肠癌提供实验依据。方法将体外培养的结肠癌HT-29细胞分成不含药血清对照组、5-FU组、高剂量四君子汤血清加5-FU组、中剂量四君子汤血清加5-FU组和低剂量四君子汤血清加5-FU组5组。将制备好的四君子汤含药血清和5-FU按所需的剂量分别加入到各组培养瓶中作用于细胞24h,应用基于酶标仪的MTT方法检测各组细胞的OD值,计算不同浓度药物处理对细胞生长的抑制率。结果四君子汤血清与5-FU联用能显著抑制结肠癌HT-29细胞的生长(均P<0.01),且高剂量四君子汤血清与5-FU合用组细胞生长抑制率显著高于5-FU单用组(P<0.01)。四君子汤血清与5-FU联用对结肠癌HT-29细胞周期的运行产生显著的影响。结论四君子汤可以增强5-FU对结肠癌HT-29细胞的生长作用和细胞周期的干扰作用。s: Objective To investigate the inhibitory effect of Sijunzi decoction ( SJZD ) combined with 5 -FU on the growth of colon cancer HT- 29 cells. Methods Human colon cancer HT- 29 cells cultured in vitro were divided five groups as follow: non-drug serum (control), 5 -FU( 501amol/L ), high-dose SJZD serum combined with 5 -FU, medium-dose SJZD serum combined with 5 -FU, low-dose SJZD serum combined with 5 -FU. The cells were treated with 5 -FU and different doses of SJZD serum respectively for 24h, the growth activity of HT- 29 cells were detected by MTT assay, and the cell cycles were tested by FCM methods. Results SJZD serum combined with 5 -FU can inhibit the growth of HT- 29 cells. Growth inhibition rate of high- dose SJ2D serum combined with 5 -FU group was significantly higher than that of 5 -FU group(P〈0.01 ), SJZD serum combined with 5 -FU affected the HT- 29 cell cycle significantly. Conclusion SJZD can enhance the effect of 5 -FU on growth inhibition and cell cycle interference of colon cancer HT- 29 ceils.
