摘 要:本研究旨在提出通过综合评价方法对耐热植酸酶进行系统评估,为耐热植酸酶性能快速判断和实际应用提供借鉴和指导。本研究以TP1、TP2、TP3三种不同的市售耐热植酸酶为对象。分别从模拟高温制粒耐热性、模拟体内活性和内源蛋白酶耐受性以及体外酶解效果四方面分析对比。然后综合考虑各阶段的酶活存留来评估对植酸磷的酶解效果。结果表明,在仅考虑耐热性时TP3最优:在仅考虑体内活性和内源蛋白酶耐受性时,TP1和TP2相对较好;在仅考虑对植酸磷的释放力时,TP1与TP2也相对较好;然而,综合考虑耐热性、体内活力和内源蛋白酶耐受性以及植酸磷释放四方面,分析发现TP1效果最优。由此可见,耐热植酸酶的评估应根据实际应用的环节和各阶段过程,形成综合有效评估耐热植酸酶应用效果的方法尤为重要,这对植酸酶的前期选择具有重要的指导意义。The aim of this study was to provide a data support for the application of thermo- stable phytase in the actual production by systematically evaluating the effect of phosphorus release in phytic acid. This study includes three aspects: the effect evaluation in high temperature granulation process, the effect evaluation of in vivo and the effect evaluation of enzymatic hydrolysis in vitro. Taking the three kinds of therrnostable phytase as objects, which were respectively marked TP1, TP2 and TP3, then evaluating the comprehensive effect on enzymatic hydrolysis of phytic acid by the changes of enzyme activities in different stages. The results showed that TP3 had the best heat resistance, TP1 and TP3 were both better than TP2 in this point; In vivo tolerance and the release of phytate phosphorus, the effect of TP1 and TP2 were equivalent, both better than TP3. In a word, the system evaluation of therrnostable phytase should be based on the actual application. It' s important to form a comprehensive and rapid assessment method of thermostable phytase, which is of important guiding significance for the practical application of thermostable phytase.
