摘  要:目的 依据穿心莲内酯和脱水穿心莲内酯的稳定性机理,结合清火栀麦片的制法,以君药穿心莲主要成分穿心莲内酯和脱水穿心莲内酯为指标,优化HPLC法测定清火栀麦片中穿心莲含量的提取条件。方法分析比较不同提取溶剂、提取方法、提取时间及提取温度对HPLC法测定穿心莲含量结果的影响。结果与其他提取溶剂相比,甲醇对穿心莲的提取效率较高,且提取液的杂质含量更低;提取中超声前是否冷浸、加冰或常温对检测结果影响不大;提取时超声1 h较超声30 min的提取效果更好;而在低温时,随着温度的上升有助于主要成分的提取,但当温度超过60℃时发生降解。结论以甲醇为溶媒,常温超声1 h时提取纯度和提取效率较优。Objective To optimize the extraction conditions of HPLC for the determination of andrographis paniculata in Qinghuo Zhimai tablets, based on the stability mechanism of andrographolide and dehydroandrographolide combined with the preparation method of Qinghuo Zhimai tablets, with the indicator of the main ingredient of herbs andrographis including andrographolide and dehydrated andrographolide. Methods The effects of different extraction solvents, ex- traction methods, extraction time and extraction temperature on the determination of andrographis paniculata by HPLC were analyzed and compared. Results Compared with other extraction solvents, the extraction efficiency of methanol to andrographis paniculata was higher, and the impurity content of the extract was lower. Whether cold dip, adding ice, room temperature or not bad little effect on the detection result. The extraction effect of ultrasound for 1 h was better than that of ultrasound for 30 min. While at low temperature, the increase of temperature was helpful to the extraction of the main components, but the degradation occurred when the temperature exceeded 60℃. Conclusion The extraction purity and extraction efficiency were better when using methanol as the solvent at room temperature for 1 h.
