摘  要:目的:研究金花茶粗提物与正丁醇提取物对二乙基亚硝胺(DEN)诱发的肝癌前病变的预防作用。方法:实验用清洁级雄性C57BL/6小鼠,将传统的Solt-farber模型加以改良,给予DEN与二乙酰氨基物(2-AAF)之后切除部分肝脏,诱发肝癌。各实验组开始即给予100mg/kg的金花茶粗提物与100mg/kg的金花茶正丁醇提取物,对照组给予羧甲基纤维素钠溶液灌胃,16周后处死小鼠。观察小鼠肝脏的病变程度,血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)、超氧化物歧化酶(SOD)、丙二醛(MDA)的含量,及肝脏组织中炎症因子IL-6、IL-1β、TNF-α、IFN-γ水平的变化。结果:与模型组比较,粗提物组与正丁醇组小鼠的肝脏病变程度明显减轻,血清中ALT、AST及MDA含量较模型低,但SOD没有变化。组织中炎症因子较模型组低,但无显著变化(P>0.05)。结论:金花茶提取物对诱发小鼠肝癌具有一定的预防作用,可能与其抗氧化作用有关。Objective:To Study the preventive effect of crude extract and n-butanol extract from Camellia chrysantha Tuyama on hepatic precancerous lesion induced by diethylnitrosamine. Methods: Laboratory C57BL/6J mice, all being male and clean grade, were administered with DEN and 2 Acetaminoflueren(2-AAF), in conjunction with partial hepatectomy by an improved Solt-farber model to induce primary carcinoma. Crude extract or n-butanol extract from Camellia chrysantha Tuyama were orally adminstrated to mice at a dosage of 100mg/kg until the mice' s euthanasia 16 weeks later. Degree of liver lesion, serum level of ALT, AST, SOD, MDA and expression of IL-6 ,IL-l[3, TNF a, IFN-7 in hepatic tissue were tested. Results:Degree of liver lesion of mice treated by crude extract or n-butanol extract from Camellia chrysantha Tuyama were significantly lightend compared with the model mice. Meanwhile, surum level of ALT, AST , MDA reduced but SOD was no change. The inflammatory cytokines in liver tissue were down but there was no significant differcence. Conclusion : To some extent, there is a preventive effect of camellia ehrysantha Tuyama for hepatic precancerous lesion,witch might be related to its antioxidantion.
