刮痧对正常大鼠局部肥大细胞功能状态的影响

摘　　要：目的:观察刮痧前后局部肥大细胞的功能状态,揭示刮痧疗法的部分作用机制。方法:正常Wistar大鼠随机分为刮痧前组、刮痧后即刻组,刮痧后30 min组、刮痧后60 min组、刮痧后90 min组,每组10只。除刮痧前组外,其余各组均给予刮痧干预,刮拭部位为大鼠背部督脉、两侧膀胱经,刮拭方法为刮法,频次为60次/分,刮拭时间30 s。各组分别于刮痧前、刮痧后即刻、30、60、90 min脱颈处死,于大鼠背部一侧膀胱经上出痧最明显处取材。分别采用甲苯胺蓝染色、免疫组化的方法观察肥大细胞数量以及脱颗粒情况、类胰蛋白酶表达情况。结果:各组肥大细胞数比较差异无显著性,P>0.05;各组肥大细胞脱颗粒率比较差异无显著性,P>0.05;各组类胰蛋白酶阳性细胞数比较差异无显著性,P>0.05。结论:刮痧后以及刮痧后的90 min内肥大细胞的功能状态并未发生明显改变,刮痧即刻未对正常大鼠局部肥大细胞产生影响,推测刮痧疗法的即刻效应与肥大细胞无关。Objective： to observe the functional status of partial mast cells before and after scraping so as to reveal the functional mechanism of Guasha therapy.Methods： The normal Wistar rats were randomly divided into pre-scraping group, instant group after scraping, 30 min group, 60 min group, and 90 min group, 10 rats each group.Except pre-scraping group, all the others were intervened with Guasha, scraping Du meridian and Bladder meridians, 60 times /min, for 30 seconds each time.The rats of each group were killed off before scraping, and instantly, 30 min, 60min and 90 min after scraping respectively, materials taken from the most obvious part of one Bladder meridian.Toluidine blue staining and Immunohistochemical tests were used to observe the number of mast cells, degranulations and tryptase expression.Results： there was no significant difference between the numbers of mast cells, P〉0.05;degranulation rates of each group had no significant difference, P〉0.05;there was no significant difference between numbers of positive tryptase, P〉0.05.Conclusion： the functional status of mast cells has no obvious changes instantly after scraping and 90 min after scraping, that is, Guasha does not affect the partial mast cells in normal rats instantly after scraping.It therefore can be predicted that the instant effect of Guasha therapy is irrelative to mast cells.
