摘 要:目的探讨2型糖尿病(T2DM)与患者KCNJ11启动子区部分Cp G位点的甲基化的相关性。方法选取15例新发单纯性2型糖尿病患者和15例健康正常对照者,用亚硫酸盐法测定KCNJ11基因启动子5’端300bp中的46个Cp G位点DNA甲基化水平,用半定量逆转录聚合酶链反应(RT-PCR)测定KCNJ11基因转录表达。结果在KCNJ11启动子区的实验区域存在多个甲基化水平增高的位点,两组比较,总甲基化比率及其m RNA表达无明显差异(t=1.0323;P>0.05),每个研究对象KCNJ11启动子区的Cp G+62、Cp G+66、Cp G+351位点甲基化比率均有升高,两组间比较无明显差异(t=0.2582,0.7821,1.1859;P>0.05),但Cp G+78、Cp G+96、Cp G+100、Cp G+104、Cp G+108、Cp G+111、Cp G+21、Cp G+129、Cp G+132、Cp G+136、Cp G+141、Cp G+168、Cp G+174、Cp G+178、Cp G+201、Cp G+210、Cp G+215、Cp G+219、Cp G+223、Cp G+233、Cp G+248、Cp G+268、Cp G+270、Cp G+272、Cp G+288、Cp G+324等26个位点的甲基化比率在2型糖尿病组的个别患者中有所升高,而在健康正常对照组却未出现甲基化。结论 2型糖尿病患者KCNJ11启动子区甲基化呈现异常,可能参与2型糖尿病的发病。Objective To investigate the correlation between methylation of partial CpG site in KCNJ11 promoter region and type 2 diabetes mellitus (T2DM). Methods 15 patients with newly diagnosed type 2 diabetes mellitus and 15 healthy controls were enrolled, DNA methylation levels of 46 CpG loci in 300bp gene promoter 5 were measured by sulfite assay in the promoter of KCNJ11 gene promoter. The transcription of KCNJ11 gene was measured by senti quantitative reverse transcription polymerase chain reaction (RT-PCR). Results In the experimental region of the KCNJ11 promoter region, multiple methylation levels were increased. The ratio of total methylation and its expression of mRNA were not significantly different between the two groups (P 〉 0.05). Each object of study of the promoter region of KCNJ11 CpG+62, CpG+66, CpG+351 methylation ratio increased, no significant difference between two groups between (P 〉 0.05), but increased the methylation ratio of CpG+78, CpG+96, CpG+100, CpG+104, CpG+108, CpG+111, CpG+21, CpG+129, CpG+132, CpG+136, CpG+141, CpG+168, CpG+174, CpG+178, CpG+201, CpG+210, CpG+215, CpG+219, CPG+223, CpG+, CPG+248, CPG+268 233, CpG+270, CPG+272, CPG+288, CPG+324 etc. 26 loci in individual patients with type 2 diabetes group, while in the normal control group without methylation. Conclusion Methylation of KCNJ11 promoter region is abnormal in type 2 diabetes mellitus, and it may be involved in the pathogenesis of type 2 diabetes mellitus.
