摘 要:目的探讨高同型半胱氨酸血症损伤神经细胞的特点。方法通过高蛋氨酸饮食在ApoE^(-/-)小鼠诱发高同型半胱氨酸血症,采用组织学,组织化学,免疫组化和原位末端脱氧核糖核酸转移酶介导的脱氧尿苷三磷酸(dUTP)标记法(TUNEL),对比观察小鼠嗅球组织结构、尼氏体变化和细胞凋亡。结果实验组血浆总同型半胱氨酸[(23.71±6.30)μmol/L]高于对照组[(6.47±2.66)μmol/L](P<0.05)。在对照组和实验组,均可见嗅球的基本组织结构,实验组嗅球组织结构未见明显的异常。Nissl体染色可见,实验组Nissl体光密度值明显低于对照组(P<0.05)。NeuN免疫组织化学染色显示,实验组NeuN阳性神经元数量与对照组差异无统计学意义(P>0.05)。实验组嗅球中TUNEL阳性颗粒细胞数量明显多于对照组(P<0.05)。结论高同型半胱氨酸血症能够导致ApoE^(-/-)小鼠嗅球神经元线粒体损伤和细胞凋亡。Objective To investigate the characteristics of neuronal cell damage induced by hyperhomocysteinemia (HHcy). Methods Hyperhomocysteinemia was induce by a high-methionine diet in ApoE-/- mice. The histology, Nissl body and apoptosis in the olfactory bulb were comparatively studied by histology, histochemistry, immunohistochemistry and terminal deoxynucleotidyl transferase- mediated dUTP nick end labeling (TUNEL). Results The total plasma homocysteine in the experimental HHcy group [(23.71±6.30) μmol/L] was significantly higher than that in the control group [(6.47±2.66) μmol/L ](P 〈 0.05). In the control group and experirnental HHcy group, the basic structure of olfactory bulb was observed, and there were no obvious histological abnormalities in the experimental HHcy group. Nissl body staining showed that the optical density of Nissl bodies in the experimental HHcy group was significantly lower than that in the control group (P 〈 0.05). As was immunohistochemistry showed that no significant difference between the numbers of NeuN-positive neuron between the experimental HHcy group and the control group (P 〉 0.05). The numbers of TUNEL-positive granule cells in the experimental HHcy group were significantly higher than those in the control group (P 〈 0.05). Conclusion The hyperhomocysteinemia may be able to induce mitochondrial damage and apoptosis in olfactory bulb neurons of ApoE-/- mice.
