摘 要:目的:研究黄芪桂枝五物汤对STZ大鼠坐骨神经Trx及Txnip表达的影响,初步探讨黄芪桂枝五物汤治疗糖尿病周围神经病变(DPN)的机制。方法:将链脲佐菌素(STZ)诱导的糖尿病大鼠随机分为模型组、给药组,另取正常大鼠为对照组。8周后,采用Western blot法及实时荧光定量PCR法测定大鼠坐骨神经中Trx与Txnip表达。结果:Western bolt法显示模型组大鼠坐骨神经Txnip表达明显高于对照组(P<0.01),给药组低于模型组(P<0.01)。实时定量PCR法显示模型组大鼠坐骨神经Txnip表达明显高于对照组(P<0.01),给药组低于模型组(P<0.01)但高于对照组。而大鼠坐骨神经Trx表达中模型组明显低于对照组(P<0.01),给药组高于模型组(P<0.01)但低于对照组。结论:DPN大鼠坐骨神经存在Txnip系统异常,黄芪桂枝五物汤治疗糖尿病周围神经病变,其作用机制与抑制Txnip表达,提高Trx表达有关。Objective: To study the influence of Huangqi Guizhi Wuwu decoction on the expression of Trx and TXnip in the sciatic nerve from diabetic rats induced by STZ,and to discuss the preliminary mechanism of Huangqi Guizhi Wuwu decoction on diabetic peripheral neuropathy( DPN). Methods: Diabetes rats induced by STZ were randomly divided into the model group and the medication group,and the normal rats were taken as the normal control group. The expressions of Txnip and Trx in the sciatic nerve were determined with Western blot and real-time PCR after eight weeks. Results:The expression of Txnip was significantly increased in the model group compared to that of the normal control group( P〈0. 01),and the medication group could obviously reduce the expression of Txnip compared to the model group by the two methods( P〈0. 01),but it was still higher than the normal control group. However,the expression of Trx was significantly lower in the model group compared to the normal control group( P〈0. 01),and it was increased by the medication compared to the model group( P〈0. 01),but was still lower than the normal control group. Conclusions:There was an abnormity of Txnip in the sciatic nerve of DPN rats,the treatment mechanism of Huangqi Guizhi Wuwu decoction for DPN may be related to the inhibition of Txnip and the enhancement of Trx.
