摘  要:为探讨运动改善心衰疾病的确切机制,采用心衰大鼠模型进行小动物跑台训练,检测PVN内微量注射PTGER3受体激动剂对交感驱动指标的影响,与假手术安静组相比,心衰安静组血浆中NE水平的升高和肾交感神经放电的增强作用发生敏感化(P<0.05).跑步运动训练明显改善心衰训练组大鼠交感驱动指标增强的敏感化作用(P<0.05).Western bolt实验结果显示:与假手术安静组相比,心衰安静组大鼠PVN内PTGER3受体蛋白和mRNA表达量发生上调(P<0.05).跑步运动训练明显下调心衰训练组大鼠PVN内PTGER3受体蛋白和mRNA表达量(P<0.05).结果提示运动训练可下调心衰大鼠PVN内PTGER3受体蛋白表达,进而抑制交感传出神经兴奋性.The goal of this study was to disclose the mechanism of exercise (ExT) in improving FFGER3 receptor mediated sympathetic outflow within the paraventricular nucleus (PVN) in rats with heart failure. Heart failure (HF) animal model was induced by coronary artery ligation and treadmill running was conducted. To examine the functional role of prostaglandin within the PVN, an agonist of PTGER3 receptor, sc- 46275 ,was microinjected into the PVN. Dose-dependent increases in plasma norepinephrine (NE), RSNA, arterial blood pressure (BP) ,and heart rate (HR) were produced in all rats. There was a significantly increase in these parameters in HF rats compared with the sham-operated rats. However, ExT significantly improved the sensitive phemomenon in HF rats. The results of western blot show that PTGER3 receptor mRNA expression levels in the PVN are also significantly increased in HF-ExT rats compared with HF-sedentary rats. Our results indicate that ExT improves PTGER3 receptor mediated sympathetic outflow within the paraventricular nucleus in rats with heart failure and the "mechanism may be involved in the downregulation of PTGER3 receptor in PVN.
