摘  要:目的 观察竹节参多糖的抗血栓活性.方法 采用大鼠腹主动脉插管取血,体外试管法测定竹节参多糖对大鼠血凝块的溶解作用;采用比浊法测定竹节参多糖对人血小板聚集的影响;采用胶原蛋白-肾上腺素诱导的血栓形成模型和血瘀证大鼠模型,观察竹节参多糖的体内抗血栓作用并探讨其作用机制.结果 体外研究时,低、中、高剂量竹节参多糖对血凝块的溶解率分别达到53.08%、57.03%和68.15%,对人血小板聚集的抑制率则分别为6.78%、29.49%和40.30%,与对照组比较差异均有统计学意义,且其血凝块溶解作用和血小板聚集抑制作用均与给药剂量呈正相关.肺血栓小鼠模型中,竹节参多糖低、中、高剂量组小鼠死亡数分别为2、2、1只,小鼠偏瘫数分别为5、4、4只,肺指数则分别为1.89%、1.71%和1.43%,均表现出显著的抗血栓作用.低、中、高剂量竹节参多糖作用于血瘀证大鼠模型后,血小板最大聚集率分别降至45.62%、41.33%和31.40%,体现了明显的抗血小板聚集作用;与模型组比较竹节参多糖低剂量组对组织型纤维溶酶激活剂(t-PA)、6-酮前列腺素F1α(6-Keto-PGF1α)的含量无明显影响,中、高剂量组可显著升高t-PA、6-Keto-PGF1α的含量,竹节参多糖各组均可显著降低大鼠体内血栓素B2(TXB2)、纤溶酶原激活物抑制因子(PAI)的含量.结论 竹节参多糖高、中、低剂量组均有显著的体外和体内抗血栓活性,其抗血栓作用可能与影响血小板功能和提高纤溶系统活性有关.Objective To investigate the antithrombotic activity of total polysaccharides from Panax japonicus C.A.Meyer.Methods In vitro,effect of polysaccharides from Panax japonicus C.A.Meyer on resolving blood clot was evaluated by tube method,and that on human platelet aggregation was by turbidimetry method.The collagen-epinephrine-induced thrombosis model in mice and blood stasis syndrome model in rats were used to study the antithrombotic activity and the antithrombotic mechanism of total polysaccharides in vivo.Results In the In vitro study,blood clot solubility of low,moderate and high dosage of polysaccharides group was 53.08%,57.03% and 68.15% respectively,and inhibition rate of platelet aggregation was 6.78%,29.49% and 40.30% respectively.All these effects had a positive correlation with dose.In collagen-epinephrine-induced thrombosis mice model,the number of deaths in low,moderate and high dose group was 2,2 and 1,respectively.The number of hemiplegia in these groups was 5,4 and 4 with the lung index showing a very significant reduction (1.89%,1.71% and 1.43% respectively).The number of deaths and hemiplegia in low dose group was 2 and 5,respectively,and the lung index showed a significant reduction.In blood stasis syndrome rat model,all polysaccharides dose groups showed significant inhibition activity of platelet aggregation.Maximum aggregation rate was decreased to 45.62%,41.33% and 31.40% respectively.Compared to the model group,there was no significant effect on Tissue-type plasminogen activator (t-PA) and 6-keto-prostaglandin F1α (6-Keto-PGF1α) in low dose group.Middle and high dose group could significantly increase the contents of t-PA and 6-Keto-PGF1α.All polysaccharides dose groups showed a significant reduction in the contents of thromboxane B2 (TXB2) and plasminogen activator inhibitor (PAI) in blood stasis syndrome model rats.Conclusion The high,middle and low doses of total polysaccharides from Panax japonicus C.A.Meyer had the significant antithrombotic activity.The antithro
