摘 要:目的观察DON毒素与氟致骨软骨损伤的联合作用机制中,对转化生长因子β2(TGF-β2)、成纤维细胞生长因子23(FGF-23)等4种骨生长发育相关的指标的影响,探讨DON毒素与氟的联合作用和茶的拮抗效应。方法 28日龄Wistar大鼠幼鼠30只,按照完全随机化方法分空白组、"DON+F"组和"DON+F+茶"组。空白组饮用超纯水,"DON+F"组饮用150mg/L NaF超纯水溶液;"DON+茶+F"组饮用由茶水溶解的150mg/L NaF水溶液;DON毒素染毒组各组按0.2μg/g DON毒素灌胃,空白组用等量超纯水灌胃,每天1次。喂养5周后,麻醉后断头取血,ELISA法测定各细胞因子。结果 ELISA结果显示,血清中TGF-β2的平均含量"DON+F"组(55.47pg/ml)显著高于对照组(29.2 pg/ml)和"DON+F+茶"组(38.4 pg/ml);FGF-23的平均含量"DON+F"组(205.49pg/ml)显著高于对照组(192.92 pg/ml)和"DON+F+茶"组(97.3pg/ml);BMP-2的平均含量"DON+F"组(15.24ng/ml)显著高于对照组(12.44ng/ml)和"DON+F+茶"组(12.9ng/ml);TNF-α的平均含量"DON+F"组(936.88pg/ml)显著高于对照组(579.54 pg/ml)和"DON+F+茶"组(652.95 pg/ml)。结果提示DON与F可导致骨软骨损伤,茶具有保护作用。结论茶可在一定程度上可减轻DON毒素和氟对骨软骨的损伤,显示了一定的拮抗作用,但不能消除DON毒素和F的致病效应。Objective To observe the effect of tea on the mechanism of bone and cartilage damage induced by DON toxin and fluoride in 4 kinds of bone growth and development, to investigate the effect of tea on the mechanism of bone damage induced by DON toxin combined with fluoride. Methods Thirty female rats were randomly divided into 3 groups: control, DON + F, and DON +F +Tea group.Control group was drunk with deionized solution, DON+F group, deionized solution of NaF was drunk with150mg/L, DON+ tea +F group was drunk 150 mg/L NaF water solution dissolved by tea water, DON + F and DON + F + Tea group, deionized solution of DON was gavage with a dose of 0.2 μg/g every day, control group was gavage with deionized solution every day.After 5 weeks, broken blood, the cytokines were determined by ELISA.Results The results of ELISA showed that the concentration of TGF-β2 in the serum of DON+F group( 55.47 pg/ml) were significantly higher than those in the control group( 29.20 pg/ml) and DON +F +Tea group( 38.4 pg/ml); The concentration of FGF-23 in the serum of DON+F group( 205.49 pg/ml) were significantly higher than those in the control group( 192.92 pg/ml) and DON +F +Tea group( 97.3 pg/ml); The concentration of BMP-2 in the serum of DON+F group( 15.24 ng/ml) were significantly higher than those in the control group( 12.44 ng/ml) and DON +F +Tea group( 12.9 ng/ml); The concentration of TNF-α in the serum of DON+F group( 936.88 pg/ml)were significantly higher than those in the control group( 579.54 pg/ml) and DON +F +Tea group( 652.95 pg/ml). The results suggest that DON and F may cause osteochondral damage, tea has a protective effect.Conclusions Tea can reduce the damage of DON toxin and fluorine on bone cartilage to a certain extent, showed some antagonism, but don't eliminate the effects of DON toxin and F.
