摘 要:目的了解宁波市2016年流行性腮腺炎病毒(Mumps virus,Mu V)基因型特征。方法采用Vero/SLAM细胞分离Mu V,对Mu V毒株小疏水蛋白(Small hydrophobic protein,SH)基因进行逆转录-聚合酶链反应扩增和序列测定,与Gen Bank中参考株序列进行比较。结果宁波市2016年共分离到5株Mu V,均为F基因型,核苷酸及氨基酸之间同源性分别为95.07%和94.74%;与F基因型参考株Z77161同源性分别为96.42%-97.09%和94.74%-96.49%;与中国疫苗株S79同源性分别为80.66%-81.52%和73.68%-75.44%;与中国其他地区F基因型之间同源性分别为95.05%-97.42%和91.23%-96.49%。结论 2016年宁波地区Mu V流行株均为F基因型,未发现SH基因变异。Objective To analyze genotype characteristic of mumps virus(Mu V) strains isolated in Ningbo,2016. Methods Mu V strains were isolated using Vero/SLAM cells. The small hydrophobic protein(SH) gene of Mu V was amplified by RT-PCR. Sequences were analyzed phylogenetically and compared with other reference strains published in Gen Bank. Results Five strains of Mu V were isolated and identified as genotype F,with a homology of 95. 07% in nucleotides(nt) and 94. 74% in amino acids(aa).The nt and aa homology were 96. 42%-97. 09% and 94. 74%-96. 49% compared with Z77161 strain,80. 66%-81. 52% and 73. 68%-75. 44% compared with China S79 vaccine strain, and 95. 05%-97. 42% and 91. 23%-96. 49% compared with other strains isolated in china,respectively. Conclusions The main strain of Mu V circulating in Ningbo is genotype F without mutations in the SH gene.
