摘 要:目的观察益气祛风通络饮对肾脏纤维化相关指标纤维连接蛋白(fibronectin,FN)、层粘连蛋白(laminin,LN)和自噬相关蛋白LC3、Beclin-1表达的影响。方法健康雄性SD大鼠40只,其中以30只为造模组,采用单次尾静脉注射阿霉素造模,造模成功后随机分为模型组、替米沙坦组及益气祛风通络饮组,每组各10只,分别予生理盐水、替米沙坦混悬液及益气祛风通络饮水煎剂灌胃治疗;以10只为空白组,予生理盐水灌胃。实验结束后于光镜下行肾组织病理学观察,采用免疫组化法检测FN、LN、LC3和Beclin-1的表达及分布。结果 (1)与模型组比较,益气祛风通络饮组大鼠在第4~8周24 h尿蛋白定量均明显下降(P<0.05);与替米沙坦组比较,两组差异无统计学意义(P>0.05)。(2)肾组织病理:与模型组比较,益气祛风通络饮组及替米沙坦组肾脏组织损伤均有不同程度的减轻。(3)肾组织中FN、LN的表达:与空白组比较,模型组肾组织中FN、LN表达水平显著升高(P<0.05);与模型组比较,益气祛风通络饮组肾组织中FN、LN表达水平显著降低(P<0.05);益气祛风通络饮组与替米沙坦组比较,肾组织中FN、LN表达水平差异无统计学意义(P>0.05)。(4)肾组织中LC3、Beclin-1的表达:与空白组比较,模型组肾组织中LC3、Beclin-1表达水平显著升高(P<0.05);与模型组比较,益气祛风通络饮组肾组织中LC3、Beclin-1表达水平显著降低(P<0.05);益气祛风通络饮与替米沙坦组比较,肾组织中LC3、Beclin-1表达水平差异无统计学意义(P>0.05)。结论益气祛风通络饮能够降低阿霉素大鼠蛋白尿,其对肾脏病理具有改善作用,可能是通过下调肾组织中FN、LN的表达水平和拮抗自噬相关蛋白Beclin-1、LC3的过度表达而实现。Objective To observe the impacts of yiqi qufeng tongluo decoction on the expressions of fibronectin(FN)and laminin(LN)relevant with renal fibrosis and LC3 and Beclin-1 relevant with autophagy.Methods Of 40 healthy male SD rats,30 rats were used in modeling group,in which,the models were prepared by single injection of Adriamycin in the vein of tail.After modeling success,a model group,a telmisartan group and yiqi qufeng tongluo decoction group were randomized,10 rats in each one,treated with lavage of physical saline,telmisartan suspension and yiqi qufeng tongluo decoction separately.The rest 10 rats were used in the blank group for lavage with physical saline.At the end of experiment,the pathological changes of renal tissue were observed under microscope.The immunohistochemical method was adopted to detect the expressions and distributions of FN,LN,LC3 and Beclin-1.Results 1.Compared with the model group at the same time phase,the 24 h urine protein was reduced apparently in 4th to 8th weeks in the rats of yiqi qufeng tongluo decoction group(P〈0.05).The difference was not significant at the same time phase as comparedwith the telmisartan group(P〉0.05).2.Pathology of renal tissue:compared with the model group,the damages of renal tissue were alleviated to different degrees in yiqi qufeng tongluo decoction group and telmisartan group.3.The expressions of FN and LN in renal tissue:compared with blank group,the expressions of FN and LN in renal tissue were increased significantly in the model group(P〈0.05).Compared with the model group,the expressions of FN and LN in the yiqi qufeng tongluo decoction group were reduced significantly(P〈0.05).In comparison between the yiqi qufeng tongluo decoction group and telmisartan group,the differences in the expressions of FN and LN were not significant(P〉0.05).4.The expressions of LC3 and Beclin-1 in renal tissue:compared with the blank group,the expressions of LC3 and Beclin-1 were increased significantly in the model group(P〈0.0
