秦艽分离有效部位对胶原诱导性关节炎的作用研究

摘　　要：目的:分离并筛选秦艽有效部位,研究其对胶原诱导性关节炎大鼠的治疗作用及作用的分子机制。方法:通过D-101大孔树脂对秦艽醇提物进行分离纯化;采用二甲苯致小鼠耳肿胀急性炎症模型观察秦艽各分离片段的抗炎活性;进一步通过胶原诱导性关节炎大鼠模型,研究秦艽10%乙醇分离片段对大鼠足肿胀度、外周血中Ⅱ型胶原抗体、TNF-α、IFN-γ和IL-4水平的影响;并利用UPLC法测定各分离片段中4种环烯醚萜类化合物的富集情况。结果:秦艽10%乙醇分离片段(生药5g/kg)显著抑制二甲苯所致小鼠耳肿胀;在胶原诱导性关节炎大鼠模型实验中,秦艽10%乙醇分离片段(生药3.5g/kg)能显著减轻关节肿胀,降低Ⅱ型胶原抗体、TNF-α和IFN-γ水平,升高IL-4水平。通过UPLC法检测发现10%乙醇分离片段中的主要成分为4种环烯醚萜类化合物,总含量为69.98%。结论:D-101大孔树脂可用于秦艽醇提物的分离纯化,10%乙醇分离片段为秦艽抗类风湿性关节炎的有效部位,通过UPLC法测定该有效部位的主要成分为环烯醚萜类化合物。Objective： To separate and screen the effective part from Gentiana, and study the treatment and molecular mechanism of the effective part on type Ⅱ collagen-induced arthritis in rats. Methods： Gentiana alcohol extract were isolated and purified by D-101 macroporous resin. The acute inflammatory models, such as xylene-indueed ear edema was used in researching the inflammatory effects of isolated fragments of Gentiana. Collagen induced arthritis rats was used to investigate the 10% ethanol fragment of Gentiana on rat foot swelling and the level of TNF-α,IFN-γand IL-4 in peripheral blood. Then UPLC method was used to detect the eluting condition of 4 kinds of iridoid compounds. Results： The 10% ethanol fragment of Gentiana （5 g/kg） could significantly inhibit ear edema in mice （P 〈 0.01 ）. In the experiment of collagen-induced arthritis （CIA） models in rats, the 10% ethanol fragment of Gentiana （3.5 g/kg） could reduce rat foot swelling（ P 〈0. 01 ）, type Ⅱ collagen antibody（ P 〈 0.01 ）, TNF-α（ P 〈 0.05 ） and IFN-γ level gradually （P 〈 0.05 ）. It also could increase IL-4 level significantly（ P 〈0.01 ）. Detected by UPLC, the main components in 10% ethanol fragment were the 4 kinds of iridoid compounds and the content is 69.98%. Conclusion： D-101 macroporous resin can be used to separate and purify the Gentiana ethanol extraction, the 10% ethanol fraction is the effective part to against rheumatoid arthritis. Iridoid compounds are the main components in 10% ethanol fraction. This paper provides a basis for the separation, quality control and treatment of rheumatoid arthritis of Gentiana.
