摘 要:目的探讨不同浓度非布司他对高糖诱导的人肾小管上皮细胞(HK-2)增殖、凋亡及胞内信号通路的影响。方法通过CCK8法和流式细胞术观察不同浓度非布司他对高糖诱导的HK-2增殖与凋亡的作用。采用蛋白免疫印迹法检测p22、p47磷酸化蛋白表达水平。结果 100μmol/L非布司他具有最佳的逆转高糖抑制HK-2细胞增殖的作用,可改善高糖诱导的HK-2细胞凋亡,降低高糖介导的HK-2的p22、p47磷酸化蛋白表达水平(P<0.01)。结论非布司他可通过抑制p22、p47磷酸化蛋白表达,改善高糖诱导的HK-2凋亡,以100μmol/L浓度效果最优。Objective To investigate effects of different concentrations of Febuxostat on cell proliferation, apoptosis and oxidation, and signal pathway of high glucose-induced human renal tubular epithelial cell. Methods Effects of different concentrations of Febuxostat on cell proliferation and apoptosis of high glucose-induced human renal tubular epithelial cell were observed by CCK8 and flow cytometry methods. Expression levels of p22 and p47 phosphorylated proteins were detected by western blot method. Results The 100 μmol/1 Febuxostat had the best effect of reversing cell proliferation inhibited by high glucose human renal tubular epithelial cell, and it could improve high glucose-induced renal tubular epithelial cell apoptosis and inhibit expression levels of p22 and p47 phosphorylated proteins of high glucose- mediated human renal tubular epithelial cell (P 〈 0. 01 ). Conclusion Febuxostat can improve high glucose-induced human renal tubular epithelial cell apoptosis by inhibiting expression levels of p22 and p47 phosphorylated proteins, and 100 umol/1 Febuxostat can achieve the best effect.
