摘　　要：目的:观察表没食子儿茶素没食子酸酯(EGCG)在同型半胱氨酸(Hcy)促血管平滑肌细胞(VSMCs)增殖中的作用,了解其对VSMCs增殖的保护作用,并初步探讨相关机制。方法:以人主动脉平滑肌细胞(HASMCs)为研究对象,分为对照组(control)、Hcy组(Hcy100μmol/L、Hcy200μmol/L、Hcy500μmol/L、Hcy1 000μmol/L)、EGCG组(EGCG5μmol/L、EGCG10μmol/L、EGCG20μmol/L)、Hcy+EGCG组(Hcy500μmol/L、Hcy500μmol/L+EGCG5μmol/L、Hcy500μmol/L+EGCG10μmol/L、Hcy500μmol/L+EGCG20μmol/L);在培养的第24 h,采用CCK-8法测定细胞增殖情况,Brd U染色法标记增殖细胞,ELISA方法测定Hcy+EGCG组中血管紧张素Ⅱ(AngⅡ)、血管紧张素1-7(Ang1-7)水平。结果:Hcy可促进HASMCs增殖,EGCG可抑制HASMCs增殖,EGCG可抑制Hcy促HASMCs的增殖作用,其发挥作用与其抑制HASMCs分泌AngⅡ、诱导HASMCs分泌Ang1-7有关。结论:EGCG可通过调节局部AngⅡ/Ang1-7平衡来抑制Hcy诱导的HASMCs增殖。Objective：To observe the protective effect of epigallocatechin gallate on homocysteine - induced human aortic smooth muscle cells（HASMCs） proliferation and discuss the mechanisms. Methods：HASMCs served as objects, which were placed in various culture media, including homocysteine, epigallocateehin gallate or the above regents for 24 h. The proliferation of HASMCs was determined by cell counting kit - 8 （ CCK - 8 ） and bromodeoxyuridine （ BrdU ） staining. The levels of angiotensinII and angiotensinl -7 were determined by ELISA. Results：Homocysteine induced the proliferation of HASMCs and epigallocatechin gallate inhibited the proliferation of HASMCs. Epigallocatechin gallate suppressed homoeysteine - induced proliferation of HASMCs, and the inhibitory effects was associated with the suppression of angiotensinⅡ secretion and the promotion of angiotensinl - 7 secretion. Conclusion： Epigallocateehin gallate inhibited homoeysteine - induced proliferation of HASMCs by adjusting local angiotensinII/angiotensinl -7 balance.
