摘 要:目的探讨红景天苷对家兔50%减体积原位肝移植实验性术后疲劳综合征(POFs)的作用及其机制。方法将36只家兔随机分为3组,每组12只。A组不手术,B、C组先采用小肠切除-吻合术建立POFs模型,造模第10天将两组肝脏进行交叉50%减体积原位移植,术后C组用10g/L红景天苷按2mL/kg剂量经尾静脉注射治疗7d;A组、B组按1mL/kg剂量尾静脉注射生理盐水。检测3组家兔造模前、肝移植术后24h和治疗第7天大脑皮质电位、外周血5-羟色胺(5-HT)和垂体生长激素(GH)水平,并检测谷丙转氨酶(ALT)、谷草转氨酶(AST)和总胆红素(TBIL)含量。结果 C组家兔治疗第7天与肝移植术后24h及B组比较,5-HT、大脑皮质电位及肝功能指标ALT、AST和TBIL均明显降低,GH明显增高,差异均有显著意义(F=50.21~102.75,q=3.98~14.02,P<0.05)。结论红景天苷可降低家兔50%减体积原位肝移植实验性POFs并促进肝功能恢复,对家兔实验性POFs有一定程度的防治作用。Objective To investigate the effect of salidroside on postoperative fatigue syndrome(POFs)in rabbits undergoing 50% reduced-size orthotopic liver transplantation. Methods A total of 36 rabbits were randomly divided into three groups,with 12 rabbits in each group.Group A was not given surgery;in groups B and C,small intestinal resection-anastomosis was performed to establish a rabbit model of POFs,and 50%reduced-size orthotopic liver transplantation was performed on day 10 after modeling.After surgery,the rabbits in group C were given tail vein injection of 10g/L salidroside at a dose of 2mL/kg for7 days,and those in groups A and B were given tail vein injection of normal saline at a dose of 1mL/kg.Cerebral cortex potentials,peripheral 5-hydroxytryptamine(5-HT)and pituitary growth hormone(PGH)levels,and levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST),and total bilirubin(TBIL)were measured before modeling,at 24 hafter liver transplantation,and on day 7of treatment. Results Group C had significant reductions in 5-HT,cerebral cortex potentials,and levels of ALT,AST,and TBIL and a significant increase in PGH at 24 hafter liver transplantation and on day 7of treatment,and compared with group B,group C had significantly lower 5-HT,cerebral cortex potentials,and levels of ALT,AST,and TBIL and a significantly higher level of PGH(F=50.21-102.75,q=3.98-14.02,P〈0.05). Conclusion In rabbits undergoing 50%reduced-size orthotopic liver transplantation,salidroside can reduce experimental POFs and promote the recovery of liver function,and therefore,it can be used for the prevention and treatment of experimental POFs in rabbits.
