摘 要:目的研究戊酸雌二醇对大鼠胎鼠睾丸与睾丸引带形态结构及功能的影响,探讨睾丸下降不全的原因。方法30只健康SD受孕大鼠随机均分为4组:对照组及低、中、高剂量组。戊酸雌二醇以剂量0.8~2.0 mg/(kg·d)灌胃作用于怀孕6~15 d SD母鼠,分娩后正常喂养雄性子代,出生60 d测量附睾、睾丸的脏器系数(附睾、睾丸重量/大鼠体重100),进行睾丸组织学切片观察生精小管的形态学变化,测量生精小管直径。结果中、高剂量组睾丸均有一定程度的下降不全,光镜下睾丸生精小管、支持细胞存在明显的发育障碍,睾丸Leydig细胞明显增生;附睾、睾丸脏器系数、睾丸生精小管直径较对照组及低剂量给药组差异有统计学意义(P<0.05)。结论雌激素可导致睾丸引带功能障碍,使睾丸下降发生异常而诱导隐睾发生;同时造成睾丸Sertoli细胞、睾丸Leydig细胞和生精细胞发育障碍及功能改变。Objective To explore the effects of estradiol valerate on the testis and testicular gubernaculum of fetal SD mice in vivo and to investigate the mechanism of estradiol valerate-induced cryptorchidism. Methods 30 pregnant SD mice were randomly and equally divided into a blank control,a low dose,a medium dose and a high dose group to receive in tragastric estradiol valerate at 0. 8- 2. 0 mg/kg. The weight of the testis,the morphology and location of the testis and gubernaculum,the organ coefficients of the testis and epididymis were obtained on PND 60 days after birth,the morphological changes of spermatogenic cells were observed by testis biopsy,and the diameter of the seminiferous tubule and epithelial height were measured. Results Under the light microscope,hypotrophy was seen in all the testis sem iniferous tubules,spermatogenic cells and sertoli cells,marked Leydig cell hyperplasia was noted in the medium and high dose group( P〈0. 05). Conclusion Estradiol valerate could cause dysfunction of the testis gubernaculum,induce cryptorchidism,and cause dysplasia and dysfunction of sertoli cells,to investigate the causes of testicular descent.
