摘  要:目的鉴定五株内生真菌并初步研究其次级代谢产物的化学成分。方法运用分子生物学手段鉴定五株内生真菌,采用硅胶柱色谱、高效液相色谱法及TLC研究其次级代谢产物的化学成分,并运用酶标法测定单体化合物的单胺氧化酶抑制活性。结果与结论 GWT-8、LRWT-26-1和LRWT-39鉴定为链格孢属真菌,GWT-7鉴定为喙枝孢属真菌,CQ-17暂时鉴定为镰孢属真菌。从其次级代谢产物中分离得到5个化合物pseurotin A1(1)、alternariol(2)、ergosterol peroxide(3)、bisdethiobis(methylthio)gliotoxin(4)、alternariol-4-methy ether(5),其中化合物3和化合物4具有一定程度的单胺氧化酶抑制活性,初质量浓度为300μg·mL^(-1)时,抑制率分别为53.82%和65.60%,IC50值分别为142μg·mL^(-1)和130μg·mL^(-1)。To identify five strains of endophytic fungi by means of molecular biology,research on the chemical composition of metabolites was carried out with silica gel column chromatography,high performance liquid chromatography and TLC,and monoamine oxidase inhibitory activity was evaluated with enzymelabeled immunosorbent assay. GWT-8,LRWT-26-1 and LRWT-39 were Alternaria sp. GWT-7 was identified as Rhinocladiella sp.,CQ-17 was identified as Fusarium sp. Five compounds[pseurotin A1( 1),alternariol( 2),ergosterol peroxide( 3),bisdethiobis( methylthio) gliotoxin( 4),alternariol-4-methy ether( 5) ]were isolated from their metabolites. Ergosterol peroxide( 3) and bisdethiobis( methylthio) gliotoxin( 4) showed moderate inhibitory activity on monoamine oxidase. The inhibition rates were 53. 82% and 65. 60%,and the IC50 were 142 μg·mL^-1 and 130 μg·mL^-1,with the initial concentration 300 μg·mL^-1.
