摘 要:目的:探讨雷帕霉素(Rapamycin,RAPA)对肾小管上皮细胞增殖和凋亡的影响。方法:体外培养肾小管上皮细胞(HK-2细胞),选择100,200,400,800 ng·m L-1雷帕霉素作用于HK-2细胞24、48、72 h。利用MTT实验分析雷帕霉素对HK-2细胞增殖的影响,并计算各浓度及不同时间的增殖抑制率优化雷帕霉素作用的浓度和时间;选择最佳的作用浓度和作用时间处理HK-2细胞,通过流式细胞分析技术,分析雷帕霉素对HK-2细胞的凋亡的影响。结果:MTT实验显示不同浓度的雷帕霉素,对增殖有抑制作用,且表现浓度依赖性和时间依赖性;雷帕霉素可以促进HK-2细胞的凋亡。结论:通过HK-2细胞模型研究,发现雷帕霉素可抑制肾小管上皮细胞增殖、促进细胞凋亡。Objective :To investigate the effect of rapamycin on the human renal tubular epithelial cell proliferation and apoptosis. Methods: HK-2 cells were cultured in vitro, and treated rapamycin with 100, 200, 400 and 800 ng ·mL^-1 for 24, 48 and 72 hours. MTF assay was used to detect the inhibition effects on HK-2 cells proliferation; and the flow cytomctry was performed to study the effects on the HK-2 cells apoptosis. Results:MTT results showed that rapamycin has inhibitory effect on the HK-2 cells proliferation depending on the concentration and time ; Flow cytometry indicated that rapamycin could promote the HK-2 ceils apoptosis. Conclusion: The cell model shows that rapamycin inhibits the renal tubular epithelial cells proliferation and promotes the renal tubular epithelial cells apoptosis.
