摘  要:目的:应用双向凝胶电泳和质谱技术研究三周轻负重游泳训练对小鼠肝线粒体蛋白质组的影响。方法:20只雄性ICR小鼠分为安静对照组(C)和游泳训练组(TB)。TB组三周轻负重游泳训练后48 h和C组小鼠均取材,提取肝线粒体蛋白质做双向凝胶电泳并分析,取差异表达10倍以上的蛋白点。质谱检测结果:C组和TB组差异蛋白表达上调5倍以上的有25个,下调5倍以上的有6个,上调10倍以上的有5个,下调10倍以上的有1个;质谱鉴定出结构明确的三种蛋白为热休克蛋白60、乙醛脱氢酶2和细胞色素b-c1复合体1。结论:三周轻负重游泳训练小鼠肝线粒体蛋白质组差异表达明显,突出地对保护线粒体和加强能量代谢电子传递过程的蛋白表达上调,这与适宜运动有利于肝脏的关系密切。[Objective]The technologies of two dimensional gel electrophoresis and mass spectrum were used to study the effects on liver mitochondria proteome of mice with three week swimming training. [Methods] Totally 20 ICR male mouse divided into control group(C) and swimming training group(TB). The TB group by light weight after 48 hours and C group were killed and the total protein of liver was determined by two dimensional gel electrophoresis separation and analyzing image and collecting data. Mass spectrum was more than 10 times Protein Spots in the two groups. [Results] There are 25 increased expression more than 5 times and 6 decreased expression.5 increased and 1 decreased more than 10 times. Protein spots of differential expression more than 10 times were selected as object protein in MS identification and three indicating proteins were identified as HSP60, ALDH and QCR1. [Conclusions] Expression of Liver mitochondria proteins in mice were differential after three week of swimming training,and increase the expression which may be the key that appropriate exercise is beneficial to the liver.
