摘 要:目的:研究藏药渣驯正丁醇部位对刀豆蛋白A(Con A)所致肝损伤的保护作用,并初步探讨其作用机制。方法:小鼠尾静脉注射Con A建立肝损伤模型,测定血清ALT、AST、TNF-α、IFN-γ及肝组织SOD、MDA、Caspase-3、Caspase-8水平,HE染色法观察肝组织病理学变化。结果:渣驯正丁醇部位能显著降低Con A所致的肝损伤小鼠血清ALT、AST、TNF-α、IFN-γ及肝组织MDA含量,升高肝组织SOD活性,降低肝组织Caspase-3、Caspase-8活化水平,减轻肝组织病理学损伤。结论:藏药渣驯正丁醇部位对Con A致小鼠肝损伤有一定的保护作用,其机制可能与抑制炎症因子释放、抗氧化活性、抗细胞凋亡有关。Objective: To investigate the protective effect and mechanism of n-butanol fraction from Tibetan medicine Brag-zhun on liver injury in mice which induced by Concanavalin A. Methods :The experimental mice were injected with Concanavalin A via the tail vein to establish the liver injury model. Serum ALT, AST, TNF-α and IFN-γ content were detected. Liver tissue SOD, MDA, Caspase-3 and Caspase-8 activities were measured. The pathological changes were detected by HE staining. Results :n-Butanol fraction from Tibet- an medicine Brag-zhun reduced the activities of ALT, AST, TNF-α, IFN-γ in serum, and hepatic MDA content, the Caspase-3 and Caspase-8 activities of liver tissues were decreased, hepatic SOD activity were increased, and alleviated the pathological injury of liver tissue which induced by Concanavalin A. Conclusion : n-Butanol fraction from Tibetan medicine Brag-zhun shows the protective effect on liver injury, which may be related to the inhibiting the release of inflammatory factors ability, antioxidative ability and anti-liver apopto- sis.
