益气化瘀清热方及其拆方对大鼠肾小球系膜细胞TGF-βRⅠmRNA及其蛋白的影响

摘　　要：目的:观察益气化瘀清热方及其拆方各类含药血清对脂多糖(lipopolysaccharide,LPS)诱导下体外培养大鼠肾小球系膜细胞(primary mesangial cells,Ms C)表达转化生长因子βI型受体(transforming growth factorβ,TGF-β-RI)mRNA及其蛋白的影响。方法:采用RT-PCR法检测各组含药血清对Ms C表达TGF-βRⅠ mRNA的影响;采用Wersten Blot法检测各组含药血清对大鼠MsC表达TGF-βRⅠ蛋白的影响。结果:(1)RT-PCR结果显示:各组含药血清均能抑制体外培养的Ms C表达TGF-βRⅠ mRNA(P<0.01),复方组作用最强,与其他含药血清组比较差异显著(P<0.01),清热组与化瘀组比较无明显差异(P>0.05),但优于益气组(P<0.01)。(2)Wersten印迹结果显示:各组含药血清均能抑制体外培养的Ms C表达TGF-βRⅠ蛋白,复方组作用最强,与其他含药血清组比较差异显著(P<0.01),清热组与化瘀组比较无明显差异(P>0.05),但优于益气组(P<0.01)。结论:益气化瘀清热方及其拆方、雷公藤多苷的含药血清均能抑制体外培养的大鼠Ms C表达TGF-βRⅠmRNA及其蛋白。复方组作用较强,清热组及化瘀组次之,益气组最弱。Objective： To observe the effect of medicated serum of Yiqi Huayu Qingre Decoction and its disassembled prescriptions on TGF-βRI mRNA and the protein expressed by rat mesangial cells cultured in vitro under the induction of lipopolysaccharide（ LPS）. Methods： Detect the effect of each medicated serum group on TGF-βRI mRNA expressed by Ms C with RT-PCR and the TGF-βRI protein by Wersten Blot. Results：（1）The results of RT-PCR： Medicated serum in groups all can restrain the expression of TGF-βRI mRNA expressed by Ms C of rats cultured in vitro（ P〈0. 01）.The effect of contained-compound group was the strongest among them and there was a significant difference compared with other medicated serum group（ P〈0. 01）. The results of contained-Qingre group and contained-Huayu group almost had no difference（ P〉0. 05）,but better than contained-Yiqi group（ P〈0. 01）.（2） The results of Wersten Blot：Medicated serum in groups all can restrain the expression of TGF-βRI protein expressed by Ms C of rats cultured in vitro（ P〈0. 01） and the influence of contained-compound group was the strongest among them and there was a significant difference compared with other medicated serum group（ P〈0. 01）. The results of contained-Qingre group and contained-Huayu group had almost no difference（ P〉0. 05）,but better than Yiqi group（ P〈0. 01）. Conclusion： Yiqi Huayu Qingre Decoction and its disassembled prescriptions as well as TW all can restrain the expression of TGF-βRI mRNA and its protein in rat mesangial cells cultured in vitro. However,the effect of contained-compound serum group is the best,and the effects of contained-Qingre serum group and contained-Huayu group are both better than that of the contained-Yiqi serum group.
