摘  要:目的:研究化瘀解毒方提取物抗胃癌细胞增殖和黏附方面的作用及机制。方法:采用高效液相色谱法(HPLC)分析化瘀解毒方提取物稳定性,化瘀解毒方提取物作用于人胃癌SGC-7901细胞株,利用MTT分析细胞增殖抑制实验,检测红细胞毒性实验,MTT快速测定法测定活性峰与Matrigel的黏附作用的影响。Western blot法检测人胃癌SGC-7901细胞中PI3K,PDK1和Akt总蛋白及其磷酸化水平的影响。结果:化瘀解毒方提取物可浓度依赖性抑制体外人胃癌SGC-7901细胞增殖、黏附,免疫印迹结果显示,化瘀解毒方提取物抑制Akt蛋白磷酸化,然而对于上游蛋白PDK1和PI3K的磷酸化无作用。结论:化瘀解毒方提取物抑制其增殖和黏附,其作用机制可能与抑制PI3K/Akt通路有关。Objective: To study the role and mechanisms of Huayu Jiedu( HYJD) on resisting gastric cancer cell proliferation and adhesion. Methods: Use high performance liquid chromatography( HPLC) to analyze the stability of HYJD extract. First,HYJD extract acts on human gastric cancer cell line SGC-7901. Analyze cell proliferation inhibition experiment by MTT to detect red blood cell toxicity experiment. MTT rapid determination method was for determining the adhesion effect of active peak and Matrigel. Detect PI3 K,PDK1 and Akt total protein and its phosphorylation level of human gastric cancer SGC-7901 cells by Western blot method. Results: HYJD extract can inhibit human gastric cancer SGC-7901 cell proliferation and adhesion on concentration dependence. Western blot analysis showed that HYJD extract inhibited Akt phosphorylation. However,it was useless for the upstream protein PDK1 and PI3 K phosphorylation.Conclusion: HYJD extracts inhibit the proliferation and adhesion of human gastric cancer SGC-7901 cell. Its mechanism may be related to the inhibition of PI3K/Akt pathway.
