摘 要:目的:观察加味四逆散对青少期应激大鼠行为学及海马、前额叶皮质中单胺类神经递质含量的影响。方法:将24只21日龄Wistar雄性大鼠随机分为空白组、模型组、氟西汀组、加味四逆散组;除空白组以外的3组予慢性温和不可预计性应激(CUMS)。造模结束后进行行为学观察,取海马及前额叶皮质,用高效液相-荧光法检测单胺类神经递质含量。结果:与空白对照组相比,模型组体质量、糖水偏爱度、旷场实验总路程、活动度、中央区路程,海马及前额叶皮质中单胺类神经递质含量均降低。加味四逆散组大鼠体质量、糖水偏爱度、旷场测试中活跃度、中央区活动路程、总路程均增加,海马及前额叶中单胺类神经递质含量均明显升高。结论:加味四逆散可能通过提高海马及前额叶皮质中单胺类神经递质的含量而改善CUMS的引起青少期大鼠抑郁样行为。Objective: To observe the effect of Jiawei Sini Powder on the behavior and the content of single amine neurotransmitter in different brain regions of adolescent stress rats. Methods: Twenty-for 21-day-old male Wistar rats were randomly divided into control group,model group,fluoxetine group,Jiawei Sini Powder group. Chronic unpredictable mild stress was applied except for the control group. HPLC with fluorescence detector was used to analyze the level of single amine neurotransmitter in different brain regions behavior observation. Results: Compared with the control group,the weight,sucrose preference,activity,the central distance and the total distance,the content of single amine neurotransmitter in the hippocampus and prefrontal cortex of the model group were lower. Compared with the control group,the weight,sucrose preference,activity,the central distance and the total distance,the content of single amine neurotransmitter in the hippocampus and prefrontal cortex of the model group were declined. Compared with the model group,the weight,sucrose preference,activity,the central distance and the total distance,the content of single amine neurotransmitter in the hippocampus and prefrontal cortex of the Jiawei Sini Powder group increased significantly. Conclution: Jiawei Sini Powder may improve adolescent stress rats behavior through increasing the content of single amine neurotransmitter in the hippocampus and prefrontal cortex.
