摘 要:【目的】为探明荔波瑶山鸡TGFβ3基因与繁殖性状的关系,寻找可作为鸡繁殖性状的遗传标记。【方法】针对TGFβ3基因设计6对引物,采用PCR测序法筛选多态位点,并利用荔波瑶山鸡母系60个个体进行繁殖性状的关联分析。【结果】荔波瑶山鸡TGFβ3基因有17个多态位点,TGFβ3基因53(T→C)、1653(C→T)和1755(A→G)位点各300日龄产蛋量、首次就巢日龄、首次就巢持续时间、首次就巢至重新开产间隔和300日龄就巢次数间呈显著或极显著相关(P<0.05或P<0.01);3343(C→T)与首次就巢持续时间、首次就巢至重新开产间隔和300日龄就巢次数均呈显著或极显著相关(P<0.05或P<0.01);3540(C→T)与开产日龄、300日龄产蛋量和首次就巢持续时间均呈显著或极显著相关(P<0.05或P<0.01);4786(C→T)和7471(G→A)分别与300日龄产蛋量和首次就巢日龄均呈显著相关(P<0.05);7263(C→T)与首次就巢持续时间呈显著相关(P<0.05)。【结论】TGFβ3基因53(T→C)、1653(C→T)、1755(A→G)、3343(C→T)、3540(C→T)、4786(C→T)、7263(C→T)和7471(G→A)与就巢性状关联显著,可作为降低就巢性提高繁殖力的辅助选择标记。[ Objective ] This study aimed to explore the correlations between TGFβ3 gene and reproduction traits in Liboyaoshan Chicken and seek genetic markets of chicken reproduction trait. [ Method] The correlations between TGFβ3 gene and reproduction traits are determined by analyzing 60 Liboyaoshan hens based on PCR technology. [ Result] There are 17 polymorphic sites in TGFβ3 gene of Liboyaoshan Chicken. 53(T→C), 1653(C→T) and 1755(A→G) sites are related to total egg number from 120 to 300 d age (EN300), age at first broodiness behavior (AFBB), duration of age at first broodiness behavior (DAFBB), interval of age at first broodiness behavior between anew laying (IAFBBA) and times of broodiness behavior from 120 to 300 d of age (TBB300) significantly or very significantly. 3343 (C→T) site is related to AFBB, IAFBBA and TBB300 significantly or very significantly. 3540(C→T) site is related to AFE, EN300 and DAFBB significantly or very significantly. 4786(C→T) and 7471 (G→A)site is significantly related to EN300 and DAFBB respectively. 7263 (C→T) site is related to DAFBB significantly. [ Conclusion ] 53 ( T→C ), 1653 ( C→T), 1755 (A→G), 3343 ( C→T), 3540 (C→T), 4786 ( C→T), 7263 (C→T) and 7471 (G→A) sites of TGFβ3 gene are significantly related to reproduction traits, which indicates that these polymorphic sites can be used as assistant selection markers for improvement of reproductive capacity of Liboyaoshan Chicken.
