摘 要:目的探讨美罗培南微透析探针在正常大鼠和脓毒症大鼠不同靶部位的回收率。方法随机将大鼠分为两组:假手术组(1只)和脓毒症组(3只)。分别将探针置入大鼠的皮肤、肌肉、血管及肺部,收集样品,测定不同部位的回收率。结果对于所研究的靶部位(肺、血管、皮肤及肌肉),正常大鼠回收率分别为30.14%±7.62%、26.20%±0.14%、25.90%±1.28%、13.95%±0.32%,脓毒症大鼠回收率分别为46.43%±1.50%、45.96%±1.78%、33.93%±0.39%、39.48%±2.38%。美罗培南在正常大鼠所研究靶部位的回收率均低于脓毒症大鼠靶部位的回收率,对于正常大鼠及脓毒症大鼠在研究的靶部位中均是肺部回收率最高。结论美罗培南为高蛋白结合率药物,肺部蛋白含量低,而美罗培南肺部吸收最少,出现肺部回收率最高的现象。通过微透析技术研究正常大鼠和脓毒症大鼠不同靶部位的回收率,可推算出不同靶部位的药物浓度。Objective Study the recovery of the meropenem micro dialysis probe in normal rats and in rats of sepsis in vivo. Method Rats randomly divided into two groups: control group (lcase)and sepsis group (3 cases), firstly, to probe into the rat skin, muscle, blood vessels and lungs, respectively, then, collect samples, and then, determination of different parts of the recovery. Result The recovery of all research target parts ( lungs, blood vessels, skin and muscles ), the normal rats were 30.14%±7.62%, 26.20%±0.14%, 25.90%±1.28%, 13.95%±0.32%. And the sepsis group were 46.43%±1.50%, 45.96%±1.78%, 33.93%± 0.39%, 39.48%±2.38%. The recovery in control group were lower than target parts of the rats of sepsis group, the recovery both in control group and sepsis group at the target of the study area are highest in lung. Conclusion Meropenem was a high protein binding rate of drug, the lung protein content was low, but the pulmonary absorption of meropenem was the least, so meropenem in the lung had the highest recovery rate. By micro dialysis technology research normal and sepsis in rats of different target parts of a recovery, and then calculate different drug concentration of the target area.
