摘 要:目的通过移植鼠源骨髓间充质干细胞(BMSCs)对SD大鼠卵巢早衰(POF)进行治疗,探讨BMSCs移植对卵巢中转化生长因子β受体Ⅱ(TGF-βRⅡ)表达的影响。方法将正常动情周期的SD雌性大鼠48只随机分为对照组、模型组(环磷酰胺致POF)、造模后自然恢复组和BMSCs移植组(尾静脉移植)。分别于建模后、BMSCs移植后取各组卵巢组织,VG染色和HE染色,光镜下观察卵巢组织形态和计数各组卵泡数。免疫组织化学法和Western blot检测各组卵巢组织TGF-βRⅡ的表达情况。结果 HE染色可见对照组有初级卵泡、次级卵泡和成熟卵泡;模型组存在闭锁卵泡,卵巢组织纤维化;自然恢复组可见闭锁卵泡和初级卵泡;BMSCs移植组可见原始卵泡、次级卵泡。BMSC移植组各级卵泡计数均高于模型组和自然恢复组(P<0.05),与对照组相比差异无统计学意义(P>0.05)。免疫组化学可见TGF-βRⅡ在模型组与自然恢复组的卵巢组织呈弱表达或未表达,BMSCs移植组卵巢组织TGF-βRⅡ阳性表达增强,Western blot检测也显示TGF-βRⅡ表达高于模型组和自然恢复组(P<0.05),与对照组相比差异无统计学意义(P>0.05)。结论 BMSCs移植可改善由化疗导致卵巢早衰的功能部分恢复,TGF-βRⅡ可能参与了移植BMSCs修复化疗后POF的过程。Objective To investigate the effect of bone marrow mesenchymal stem cells(BMSCs)on the expression of transforming growth factor βreceptor Ⅱ(TGF-βRⅡ)in ovarian premature senescence(POF) of SD rats.Methods48 SD female rats with normal estrous cycle were randomLy divided into control group, model group,natural recovery group and BMSCs transplantation group. The morphological changes of ovarian tissues were observed by light microscopy and VG staining and HE staining. The number of follicles in each group was observed after BMSCs transplantation. Immunohistochemistry and western blot were used to detect the expression of TGF-βRⅡin ovarian tissue of each group.ResultsBMSCs transplantation group showed secondary follicles and the model group had atresia follicles while ovarian fibrosis,the natural recovery group showed atresia follicles and primary follicles. Follicles presented in control group at all levels. The follicle counts at all levels in the BMSC transplantation group was significantly higher than those in the model group and the natural recovery group and it was significantly lower than that of the control group. Immunohistochemical staining showed that TGF-βRⅡwas not expressed in the ovarian tissue of the model group and the natural recovery group while the expression of TGF-βRⅡwas positive in the ovarian tissue after BMSCs transplantation. Western blot analysis showed that the protein expression of TGF-βRⅡin BMSCs transplantation group was significantly higher than those in model group and natural recovery group(P〈0.05)and there was no significant difference compared with control group(P〉0.05).ConclusionsBMSCs could improve the ovarian failure of ovarian premature senescence caused by chemotherapy,and the expression of TGF-βRⅡin POF after BMSCs transplantation was increased,which indicated that TGF-βRⅡin TGF-βsignaling pathway was involved in the treatment f POF.
