摘 要:目的:探讨大株红景天对大鼠心肌缺血再灌注损伤(MIRI)的保护作用及机制。方法:40只SD雄性大鼠随机分为正常组、假手术组、MIRI模型组、红景天预处理组,后2组结扎大鼠冠状动脉左前降支,造成缺血30min后再灌注120min,构建大鼠MIRI模型。模型组结扎前予以静脉注射氯化钠注射液,红景天预处理组予以静脉注射大株红景天注射液。测定各组血清炎症因子水平、大鼠心肌梗死面积以及心肌细胞caspase-3蛋白表达情况,评价大鼠心肌受损程度。结果:大株红景天预处理组大鼠心肌梗死面积、炎症因子水平、caspase-3蛋白表达较MIRI模型组明显降低。结论:大株红景天能明显减轻MIRI大鼠心肌损伤程度,其机制可能与抑制炎症因子释放及降低caspase-3蛋白表达有关。Objective:To evaluate the protective effects and the mechanism of rhodilla rosea on myocardial ischemia reperfusion injury (MIRI) in rats.Methods. Fourty male SD rats were randomly divided into normal group, control group, MIRI model group, rhodilla rosea pretreatment group.Left anterior descending coronary artery of rats in latter two groups was ligated by ischemia 30 min then reperfusion 120 min and the rat MIRI model was constructed. MIRI model group was given sodium chloride and rhodilla rosea group was given rhodilla rosea for intravenous administration. The serum levels of inflammatory factors, the rat myocardial infarction area and caspase--3 protein expression were measured, and the myocardial damage degree was evaluated. Results: The myocardial infarction area, levels of inflammatory, caspase--3 protein expression in rhodilla rosea pretreatment group were significantly lower than those in MIRI model group. Conclusion. Rhodilla rosea can obviously lighten MIRI rat myocardial damage,its mechanism may be related to inhibiting inflammatory cytokines release and reduce caspase--3 protein expression.
