经皮扩新术治疗实验性骨不连时BMP-2与VEGF表达及其相关性研究

摘　　要：目的观察经皮扩新术治疗实验性骨不连过程中BMP-2和VEGF的表达情况,探究该术式对两种因子表达的影响及其相关性。方法选用健康成年家兔60只,制作双前肢桡骨骨不连模型。将形成骨不连的48只兔,右前肢为实验侧,左前肢为对照侧。实验侧实施经皮扩新术,对照侧不做任何处理。于术后24、72 h、1、3、5、8周分别对两侧的放射学检查、大体标本观察、BMP-2及VEGF免疫组化染色的结果进行对比。结果经皮扩新术处理后的骨不连端表现出典型的炎症反应和修复重建过程。观察统计分析后认为实验侧和对照侧相比,BMP-2及VEGF表达的差异具有统计学意义,且两者表达呈正相关。结论经皮扩新术能够重新启动或激发骨不连时BMP-2和VEGF的表达,而且VEGF可能在一定程度上促进了BMP-2的表达。Objective To observe the expression of bone morphogenetic protein-2（BMP-2）and vascular endothelial growth factor（VEGF）during Percutaneous fracture site fresh traumas creation operation treating experimental nonunion,and to investigate the effect of the second fracture on the expression of BMP-2 and VEGF in fracture end.Methods 60 adult rabbits were selected,of which bone non-union models were made in the two radiuses of 48 rabbits.The right forelegs of the 48 rabbits with bone non-union models were prepared as experimental sides,and the left forelegs as control sides.The experimental sides were treated by Percutaneous fracture site fresh traumas creation operation.The control sides were not treated.The contrast observation of two sides were made by means of radiological examination,anatomy and immunohistochemistry staining 24 hours,72 hours,1 week,3 weeks,5 weeks and 8 weeks after operation.Results The fracture ends which were treated by Percutaneous fracture site fresh traumas creation operation reacted with typical inflammation and fracture rebuilding process.The expressions of BMP-2 and VEGF between the experimental side with the control side were significantly different.There was positive correlation between BMP-2 and VEGF by linear correlation analysis.Conclusion Percutaneous fracture site fresh traumas creation operation can re-initiate or stimulate the expression of endogenous BMP-2 and VEGF in nonunion region,which are closely correlated.To some extent,VEGF promoted the expression of BMP-2.
