摘  要:目的应用基因芯片技术,筛选可用于诊断、预测脓毒症预后的血清miRNA及其相关靶基因。方法脓毒症组:我院呼吸与危重症医学科2015年01月到2015年12月收治的汉族脓毒症患者32例;正常对照组:本单位健康体检者32例。收集2组的外周血液标本,鉴定总RNA的质量后与人类miRNA探针的基因芯片进行杂交并分析。结果脓毒症组和正常对照组中差异表达的miRNA有305个,其中在脓毒症组中上调miRNA有129个,下调miRNA有176个,上调基因中差异表达10倍以上的共有5个,分别是:miRNA-6778-3p、miRNA-6514-3p、miRNA-3591-3p、miRNA-4446-5p和miRNA-34b-3p;下调基因中差异表达10倍以上的有1个是miRNA-625-5p,在差异表达5倍以上的有5个,分别是miRNA-6756-5p、miRNA-505-5p、miRNA-130b-3p、miRNA-1914-3p和miRNA-501-5p。结论通过初步的基因芯片分析发现本组脓毒症患者异常表达的miRNA有305个,其中显著差异表达的miRNA有18个。Objective To detect and confirm the altered miRNA expression in sepsis by miRNA microarray. Methods The study selected 32 patients with sepsis from January 2015 to December 2015 as the sepsis group and 32 healthy people as the normal control group. The peripheral blood and general information were collected. The total RNA of blood samples were extracted. The quality of total RNA was determined. Total RNA samples were hybridizat- ed with miRNA probe of human gene chip. Results 305 miRNAs were detected to be significantly regulated between the sepsis group and the normal control group, 129 miRNAs were up-regulated and 176 miRNAs were down-regulated in the sepsis group. The top five up-regulated miRNAs included miRNA-6778-3p, miRNA-6514-3p, miRNA-3591- 3p, miRNA^l446-5p and miRNA-34b-3p. MiRNA-625-5p was found to be more than tenfold alterations of down-reg-ulated miRNAs. The top five down-regulated miRNAs included miRNA-6756-5p, miRNA-505-5p, miRNA-130b-3p, miRNA-1914-3p and miRNA-501-5p. Conclusion Through the analysis of miRNA microarray, 305 miRNAs are found to be expressed abnormally in patients with sepsis, 18 of which show significant differences.
