摘 要:在山东日照地区同时采集大豆[Glycine max(L.)Merr]、菜豆(Phaseolus vulgaris L.)根瘤,分离纯化,保藏根瘤菌共54株,通过提取DNA,PCR recA基因,通过Blast比对及recA基因的系统进化分析,研究日照地区大豆与菜豆根瘤菌遗传多样性。结果表明,在90%以上的相似水平上,日照大豆中的根瘤菌分布于根瘤菌属、慢生根瘤菌属、伯克霍尔德菌属3大属,圆明慢生根瘤菌、大豆慢生根瘤菌、菜豆根瘤菌、豌豆根瘤菌4个种,3个未定种。日照菜豆中的根瘤菌全部分布于根瘤菌属,菜豆根瘤菌、豌豆根瘤菌、赤小豆根瘤菌3个种,2个未定种,其中一个进化距离较远,可能为潜在新种。证明了日照大豆、菜豆根瘤菌丰富的遗传多样性,且日照大豆根瘤菌多样性大于日照菜豆。The soybean [Glycine max(L.) Merr] and kidney bean(Phaseolus vulgaris L.) rhizobia were collected simultaneously,and a total of 54 rhizobia were separated, purified and then preserved. After the extraction of total DNA,the recA gene was amplified using PCR and the obtained sequences were blasted with those selected from genbank,and phylogenetic analysis was then done. The genetic diversity of soybean and kidney bean rhizobia from Rizhao was studied. At a similarity of more than 90%, soybean rhizobia from Rizhao were identified as Bradyrhizobium yuanmingense,Bradyrhizobium japonicum,Rhizobium phaseoli, Rhizobium leguminosarum,and other three undetermined species. They were affiliated with Rhizobium,Bradyrhizobium and Burkholderia. The kidney bean rhizobia from Rizhao were identified as Rhizobium phaseoli,Rhizobium leguminosarum,Adzuki bean rhizobia,and other two undetermined species, among which one species showing a remote evolutionary distance from the others was probably a potential new species. The results proved the high genetic diversity of soybean and kidney rhizobia from Rizhao and revealed that the soybean rhizobia were more diverse than the kidney bean rhizobia.
