摘 要:为揭示脂肪分化相关蛋白基因(ADFP)、脂肪酸转运蛋白1基因(FATP1)和载脂蛋白B基因(Apo B)表达量变化对家禽脂肪沉积的影响,实验采用SYBR GreenΙ荧光定量法,首次测定了盐津乌骨鸡28、49、70、91、112 d皮脂、腹脂、肝脏和胸肌组织中ADFP、FATP1和Apo B基因的相对表达量。结果表明:ADFP基因在28 d各组织中表达量为肌肉>腹脂>皮脂>肝脏,且胸肌中表达量显著高于其他3种组织(P<0.05),在其他4个时间点各组织间表达量差异不显著(P>0.05);FATP1基因在49 d各组织中表达量为胸肌>腹脂>肝脏>皮脂,且胸肌中表达量显著高于皮脂(P<0.05),在其他4个时间点各组织间表达量差异不显著(P>0.05);Apo B基因在28、49、70、91、112 d 5个时间点各组织间表达量差异不显著(P>0.05),但在肝脏中的表达量高于其他组织。3个基因表达量变化可能在脂肪沉积过程中发挥调控作用。In order to discover the influence that adipose differentiation-related protein gene (ADFP), fatty acid transportprotein 1 (FATP1) and apolipoprotein B (ApoB) acted on fat traits of Yanjin Black-bone chickens, we adopted the SRBRGreen I Real-Time PCR method to test the relative quantity of ADFP, FATP1 and ApoB gene in subcutaneous fat, abdominalfat ,liver and chest muscle of Yanjin Black-bone chickens at 5 growth points, including 28 d, 49 d, 70 d, 91 d and 112 d. Theresults showed that the expression of the ADFP gene in 28 d is for chest muscle 〉 abdominal fat 〉 subcutaneous fat 〉 liver,and the expression of chest muscle is significantly higher than the expression of the other three tissues (P 〈 0.05), and therewas not significant in the other four time points (P〉0.05). The expression of the FATPI genes of all the tissues in 49 d showthat chest muscle 〉 abdominal fat 〉 subcutaneous fat 〉 liver, and the expression of the chest muscle is significantly higherthan the expression of the subcutaneous fat (P 〈 0.05), and there was not significant in the other four time points (P〉0.05). Thedifferrence of the expression of the ApoB gene was not significant between the tissues in the 5 time points, and the expressionof the ApoB gene in liver is significantly higher than the expression of the other tissues. The change of the three genes in thefour tissues had no significant correlation with the traits include live weight, subcutaneous fat thickness, comb weight (P 〉 0.05).The change of the quantity expression of the three genes may play a role of regulation in the process of fat deposits.
