摘 要:为筛选一株驯化啤酒酵母,并对发酵罐扩大培养工艺进行优化,为今后大规模生产提供试验数据。于麦芽汁培养基分离挑选某株具有较好生长力的酵母菌,进行形态学、生化及分子生物学鉴定。应用正交试验设计,从摇瓶温度、pH、取样时间及发酵罐的转速、装液量、接种量等方面进行优化。结果表明,驯化酵母菌株形态学、生化鉴定证明该菌株属于酵母属;发酵罐扩大培养最佳工艺为:转速200r/min,接种量50mL/L,装液量4.5L,此条件下酵母活菌含量为2.8×10~8 CFU/mL。驯化前后表现一致,没有发生变异。优化啤酒酵母菌的发酵罐扩大培养工艺,为下一步规模化生产提供理论参考。To screen a domesticated brewer's yeast and to optimize the fermentation process, and to provide experimental data for future mass production. Yeast with good growth ability was isolated and selected from the wort culture medium for morphological, biochemical and molecular biology identification. The or-thogonal experiment design was used to optimize the temperature, pH, sampling time and speed of the fer-mentation tank, liquid volume and inoculation amount. The optimum culture was as follows: the speed was 200 r/min, the inoculation amount was 50 mL/L and the volume of liquid was 4. 5 L. The content of live yeasts was 2. 8 × 10^8 CFU/mL. The performance was the same before and after domestication. Optimiza-tion of the fermentation technology of beer yeast fermentation tank expansion process provided a theoreti-cal reference for large-scale production.
