摘  要:目的:观察替普瑞酮对糖尿病肾病(diabetic nephropathy,DN)大鼠热休克蛋白72(heat shock protein 72,HSP72)表达的诱导及其对尿蛋白和血肌酐的影响。方法:将大鼠随机分为对照组、模型组和干预组,采用左肾切除+腹腔注射链脲佐菌素方法建立DN大鼠模型,干预组每日给予替普瑞酮灌胃,对照组和模型组予等体积溶媒灌胃。分别于0周(灌胃前)、6周、12周检测各组大鼠血糖、24 h尿蛋白和血肌酐水平。12周处死大鼠后,肾组织切片行过碘酸-希夫氏(PAS)染色,免疫组织化学法检测肾组织HSP72的表达。结果:灌胃12周后,PAS染色显示对照组基底膜及细胞外基质正常,模型组肾组织基底膜增厚、细胞外基质增多,干预组较模型组细胞外基质沉积减少;免疫组化结果显示干预组肾组织可检测到HSP72的表达,对照组和模型组均未检测到HSP72的表达。与模型组相比,干预组大鼠24 h尿蛋白和血肌酐明显降低(P<0.05),血糖水平差异无统计学意义。结论:替普瑞酮灌胃可诱导DN大鼠肾组织表达HSP72,并可显著降低其尿蛋白和血肌酐水平。Objective: To investigate whether geranylgeranylacetone( GGA) could induce diabetic nephrotic rats to express heat shock protein 72(HSP72) , and its effect on 24-hour urinary protein, and ser-um creatinine in diabetic nephropathy rats. Methods : The rats were randomly divided into normal control group, diabetic nephropathy group and intervention group. Left nephrectomy and streptozotocin abdomi-nal injection were carried out to establish the rat model of diabetic nephropathy. GGA was given by gavage each day for 12 weeks for the rats of intervention group and the other two groups were given equal volume of the solvent. The expression of HSP72 in the kidney tissue was detected by immunohistochemi- cal staining; and the levels of blood glucose, 24-hour urinary protein, and serum creatinine were measured for all groups at the beginning of gavage, 6 weeks and 12 weeks after the gavage, respectively. Results : When GGA had been gavaged for 12 weeks, the expression of HSP72 was detected by immunohis- tochemical staining in the intervention group, while the other two groups wasnrt in kidney tissues. The PAS staining showed that the basal membrane and the extracellular matrix were normal in control group, but the basal membrane was thickening and the extracellular matrix were increased in the model group.The extracellular matrix deposition in intervention group was less than that of the model group. Compared with the model group, the intervention group had significantly lower levels of 24-hour urinary protein and serum creatinine( P 〈 0.05 ) , but there was no statistical difference in the levels of blood glucose. Conclusion: GGA could induce the rat kidney to express HSP72, and significantly reduce the levels of urine protein and serum creatinine in rats with diabetic nephropathy.
