摘  要:目的:探讨低强度脉冲超声(low-intensity pulsed ultrasound,LIPUS)对大鼠正畸牙移动过程中牙槽骨改建中血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的影响。方法:健康6~8周雄性SD大鼠30只,采用左右对照实验,分为单纯加力组(对照组)以及LIPUS+加力组(实验组),两组各又完全随机分为1、3、5、7、14 d 5个亚组,每组6只大鼠。建立正畸牙移动动物模型,每只大鼠实验侧加力后第2天行LIPUS刺激(频率1.5 MHz、强度30 m W/cm^2、脉冲宽度200μs、重复频率1 kHz),20 min/(次·d),对照侧行不开功率的假刺激,于相应时间点处死实验鼠取上颌第一磨牙及周围牙槽骨用于免疫组织化学染色和TRAP染色。结果:免疫组织化学染色结果显示实验组VEGF的表达均高于对照组,其差异具有统计学意义(P=0.000);TRAP染色结果显示:实验组压力侧破骨细胞数均高于对照组,差异具有统计学意义(P=0.000)。结论:LIPUS能有效促进牙槽骨改建过程中VEGF和破骨细胞的表达,其可能通过增加VEGF的表达促进正畸牙移动过程中的牙槽骨改建。Objective:To investigate the effect of low-intensity pulsed ultrasound(LIPUS)therapy on the change of vascular endothelial growth factor(VEGF)in the process of the alveolar bone remodeling. Methods:Thirty male sprague-dawley(SD)rats were randomly divided into orthodontics group(control group)and LIPUS+orthodontics group(experimental group). From the second day following animal models of orthodontic teeth movement,the right maxillary took LIPUS(frequency 1.5 MHz,intensity 30 m W/cm^2,pulsewidth 200 μs,repetition frequency 1 kHz)by 20 minutes per day,and the left maxillary took sham LIPUS treatment. The samples were collected on the 1st day,3rd day,5th day,7th day and 14th day respectively. Immunohistochemistry and TRAP staining at each time point were performed to evaluate the effects of LIPUS on the change of VEGF and osteoclasts in the process of the alveolar bone remodeling. Results:The expressions of VEGF in the experimental groups were significantly higher than those in the control groups(P=0.000)and the numbers of osteoclast in the experimental groups were significantly higher than those in the control groups(P=0.000). Conclusion:LIPUS can effectively increase the expression of VEGF to promote alveolar bone remodeling.
