摘 要:为了研究袋式青贮玉米与窖式青贮玉米微生物群集的差异,同时分析与发酵产物中1,2-丙二醇相关的微生物,试验采用全株玉米进行袋式青贮以及窖式青贮。袋式青贮在3,7,14,28,56,120天开封,采样;窖式青贮选择3个500 t级别的半地上式青贮窖,贮藏120 d后每个青贮窖选择8个采样点进行采样。使用聚合酶链式反应-变性梯度凝胶电泳(PCR-DGGE)方法检测微生物区系并进行聚类分析。结果表明:袋式青贮玉米发酵过程中微生物区系无变化,在其中检测到类肠膜魏斯氏菌(W.paramesenteroides)、魏斯氏菌(W.confusa)、肺炎克雷伯菌(K.pneumoniae)、植物乳杆菌(L.plantarum)、短乳杆菌(L.brevis)和乳酸乳杆菌(L.lactis)。青贮窖不同位置微生物菌群具有一定差异,耐酸乳杆菌(L.acetotolerans)、植物乳杆菌(L.plantarum)和布氏乳杆菌(L.buchneri)为3个青贮窖共通菌,同时还检测到了屎肠球菌(E.faecium)、巴氏醋杆菌(A.pasteurianus)、罗伊氏乳杆菌(L.reuteri)、面包乳杆菌(L.panis)、嗜麦芽窄食单胞菌(S.maltophilia)和类肠膜魏斯氏菌(W.Paramesenteroides)。说明袋式青贮微生物菌群与窖式青贮微生物菌群具有一定差异,窖式青贮中检测到了一些在袋式青贮中不常见的乳酸菌;青贮窖中1,2-丙二醇的生成可能与耐酸乳杆菌(L.acetotolerans)、巴氏醋杆菌(A.pasteurianus)、罗伊氏乳杆菌(L.reuteri)、面包乳杆菌(L.panis)和布氏乳杆菌(L.buchneri)有关。This experiment was conducted to monitor variations in the bacterial community of maize silage stored in laboratory silos and bunker silos. And 1, 2 - propanediol related microorganism was analyzed. Whole crop corn was stored in laboratory silos and bunker silos. Laboratory silos were stored for 3, 7, 14, 28, 56, and 120 d, and the bacterial community was determined. Bunker silos were stored in 3 semi ground bunker silos for 120 days in the condition of 500 t class, eight samples were collected from each bunker silo. PCR - DGGE was used to deter- mine and analyze the microflora. The results showed that no distinctive changes due to prolonged ensiling were detected in the bacterial community in laboratory silos. Throughout the ensiling, W. paramesenteroides, W. confusa, and K. pneumoniae were present in addition to L. plantarum, L. brevis, and L. lactis. The bacterial community manifested variations among locations of the bunker silos. L. acetotolerans,L, plantarum and L. buchneri were the common bacteria in 3 bunkers. E. faecium ,A. pasteurianus ,L. reuteri ,L. panis ,S. maltophilia and W. parames- enteroides were detected. It was concluded that differences of bacterias were seen between bunker silos and laboratory silos. Several nonconventional silage bacteria were also detected in bunker silos. The production of 1,2 - propanediol may be associated with L. acetotolerans ,A. pasteurianus , L. reuteri , L. panis and L. buchneri.
