摘 要:目的研究五味子各萃取物抗氧化能力及抑制乙酰胆碱酯酶活性。方法五味子药材用乙醇加热回流提取后,分别用二氯甲烷、乙酸乙酯、正丁醇和水进行萃取得各组分,采用改良的Ellman方法测定五味子各组分抑制乙酰胆碱酯酶活性,并考察各组分还原能力、清除DPPH自由基作用、清除羟基自由基作用和抗脂质过氧化作用来评价抗氧化作用。结果乙酸乙酯组分(1 mg/ml)对乙酰胆碱酯酶的抑制率为(95.93±0.22)%是所有组分最高,强于阳性对照多奈哌齐(91.97±0.18)%,乙酸乙酯组分DPPH自由基清除率IC_(50)=(0.62±0.01)mg/ml和羟基自由基清除率IC_(50)=(0.07±0.01)mg/ml在所有组分中作用最强。结论乙酸乙酯组分表现出良好的抗氧化作用和抑制乙酰胆碱酯酶活性的能力,为进一步研究五味子中抗氧化及抑制乙酰胆碱酯酶活性成分提供理论依据。Objective To investigate the antioxidant and acetylcholinesterase inhibitory activities of extracts from Schisandra chinensis . Methods Schisandra chinensis is extracted by ethanol heating reflux, respectively with dichloromethane, ethyl acetate, n-butanol and water extracted components. Ellman method is used to determine the acetylcholinesterase inhibitory activities, and evaluated antioxidanation by investigated reducing capacity, scavenging actiity of DPPH free radical, scavenging actiity of hydroxyl free radical and anti- lipid peroxidation. Results The acetylcholinesterase inhibitory rates of ethyl acetate component(1 mg/ml) is (95.93±0.22)%, better than donepezil of positive control (91.97±0.18)%, is the highest in all components. The scavenging rate of DPPH free radical IC50 = (0162 ±0.01) mg/ml and hydroxyl free radical IC50 =(0.07±0.01) mg/ ml of ethyl acetate component is the highest in all components. Conclusion Ethyl acetate fraction of Schisandra chinensis can be showed good antioxidant and acetylcholinesterase inhibitory activities, the results for the further study of antioxidant activity and acetylcholinesterase inhibitory activity of Schisandra chinensis provide a theoretical basis.
