摘  要:目的:研究苦碟子注射液对氧糖剥夺损伤C6胶质细胞凋亡与自噬的调节作用,明确其对氧糖剥夺损伤胶质细胞的保护作用。方法:采用氧糖剥夺/复氧(OGD/R)的方法建立C6胶质细胞损伤模型,按随机数字表法分为空白对照组、氧糖剥夺/复氧模型组、苦碟子注射液组、自噬抑制剂(3-MA)组4组,Annexin V FITC/PI双染流式细胞仪检测细胞凋亡率,免疫荧光染色法观察活化caspase-3表达,western blotting法检测LC3Ⅱ蛋白表达。结果:氧糖剥夺/复氧损伤后LC3Ⅱ蛋白表达增加,细胞凋亡率升高,苦碟子注射液能够显著降低C6胶质细胞凋亡率,增加LC3Ⅱ蛋白高表达。结论:苦碟子注射液能够降低C6胶质细胞凋亡率,进一步激活细胞自噬而发挥保护作用。Objectives: To study the regulatory effects of Kudiezi injection on the apoptosis and autophagy of C6 glial cells,which are exposed oxygen glucose deprivation/reoxygenation injury,and clarify its neuroprotective effect on glial cells damaged by oxygen and glucose deprivation. Methods: C6 glia cells were exposed to oxygen-glucose deprivation( OGD/R) stress. The cells were randomly divided into four groups: control group,OGD/R group,Kudiezi injection group and autophagy inhibit( 3-MA) group. Apoptosis and cleaved caspase-3 were detected by Annexin V FITC/PI double staining and immunofluorescence staining. The protein expression of LCⅡ3 was detected by western blotting. Results:Oxygen-glucose deprivation/reoxygenation injury C6 cells activated LCⅡ3 protein expression increased,apoptosis increased significantly. Kudiezi injection could further activate autophagy and increase the protein expression of LCⅡ3,while reduce the rate of apoptosis. Conclusons: Kudiezi injection can reduce the apoptosis,and further activate autophagy,and plays a protective role in C6 glia cells.
