摘 要:目的研究小檗碱在常氧条件下和低氧条件下对大鼠肺三级小动脉舒缩活动的影响作用。方法分离大鼠三级肺小动脉,常氧条件下用1μmol/L苯肾上腺素(PE)预收缩血管环,累积加入不同浓度(10^(-9)—10^(-3)mol/L)的小檗碱,检测常氧条件下小檗碱对PE预收缩的血管环舒缩活动变化,计算小檗碱舒张血管效应的半数有效剂量EC50。低氧条件下10μmol/L小檗碱预孵育血管环20 min,1μmol/L PE预收缩血管环,血管环张力上升到平台期后,立即低氧,记录小檗碱在低氧条件下对PE预收缩的血管环张力变化。结果常氧条件下低浓度(10^(-9)—10^(-4)mol/L)的小檗碱以浓度依赖方式舒张血管环,其舒张效应的EC50为7.87±1.826μmol/L;高浓度(10^(-3)mol/L)的小檗碱诱导血管环收缩。低氧条件下肺小动脉环呈双相收缩变化。用10μmol/L小檗碱预孵育肺小动脉环20 min后,再经1μmol/L PE预收缩血管环和低氧刺激,低氧诱导肺小动脉环首先出现的瞬时收缩和后期出现的持续性收缩消失,血管被逆转为舒张,且出现较长时间,舒张幅度显著增加(P<0.01)。结论小檗碱可以显著抑制低氧性肺血管收缩,血管被逆转为舒张。Objetive To explore the protective effects of berberine on hypoxic pulmonary vasoconstriction. Methods Under normoxic condition, the cumulative dosing of berberine (10-9--10-3 mol/L) was added to organ chambers, in which the endothelium-intact pulmonary artery rings (PAs)were isolated and suspended, and precontracted by phenylephrine (1 μmol/L); then the changes of relaxation were recorded. Under hypoxic condition, the endothelium-intact PAs suspended in the organ chambers, were precontracted by 1 μmol/L phen- ylephrine, and then stimulated by hypoxia for 60 min to measure the contractive force in the absence or pres- ence of berberine (10 μmol/L). Results A low concentration of berberine (10-9---10-4 mol/L) could vasodilate PAs rings in a concentration-dependent manner under normoxic condition. Moreover, berberine (10 μmol/L) also removed hypoxia-induced early contraction, extended the followed maximum vasorelaxation, receded the phase Ⅱ vasoconstriction and reversed it to sustained relaxation. Conclusion Berberine could reduce hypoxic pulmonary vasoconstriction and reversed it to sustained relaxation. Conclusively, berberine may present avery potential therapy for HPV.
