寿胎丸含药血清对自然流产患者滋养层细胞凋亡相关蛋白表达的影响

摘　　要：目的探讨寿胎丸防治自然流产的可能作用机制。方法体外分离培养自然流产患者滋养层细胞,并用不同浓度(5%、10%、20%)寿胎丸含药血清干预24h(寿胎丸含药血清组),并以空白血清及地屈孕酮含药血清分别作为空白对照(空白血清组)及阳性对照(地屈孕酮含药血清组),每个浓度3个复孔。检测各组不同药物浓度血清干预后滋养层细胞总凋亡比例及细胞凋亡相关蛋白Bcl-2、Bax蛋白表达水平。结果与空白血清组比较,寿胎丸含药血清组和地屈孕酮含药血清组各浓度药物血清干预后能降低滋养层细胞总凋亡比例,上调Bcl-2蛋白水平,在浓度为10%和20%的药物血清干预后下调Bax蛋白水平(P<0.05),并且呈一定的剂量依赖性(P<0.05)。结论寿胎丸含药血清呈剂量依赖性减少自然流产患者滋养层细胞凋亡,可能与上调Bcl-2蛋白、下调Bax蛋白有关。Objective To explore the possible functional mechanism of Shoutai Wan（ 寿胎丸） in treating and preventing spontaneous abortion. Methods Trophocytes of patients with spontaneous abortion were isolated and cultured in vitro. Some of the trophocytes were intervened with serum containing Shoutai Wan in different concentration（ 5%,10% and 20%） respectively for 24 h（ as the serum containing Shoutai Wan group）,others were treated with blank serum（ as the blank serum group） or with serum containing dydrogesterone（ as the positive control group）,with 3 repetitive wells for each concentration. the total proportion of trophocytes apoptosis and the expression of apoptosis related proteins Bcl-2 and Bax were measured after being intervened with serum containing different medical concentrations. Results Compared with the blank serum group,the total proportion of trophocytes apoptosis decreased and Bcl-2 protein level increased in the serum containing Shoutai Wan group and the serum containing dydrogestrone group after intervention with drug serum in different concentration. Bax protein levels were down-regulated after intervention with 10% and 20% of drug serum（ P 0. 05）,and the down-regulation was dose-dependent（ P 0. 05）. Conclusion Shoutai Wan could reduce trophocytes apoptosis in patients with spontaneous abortion in a dose-dependent manner,which may be related with Bcl-2 protein up-regulation and Bax protein down-regulation.
