摘  要:目的观察红花水煎液对博莱霉素诱导的系统性硬化症(SSc)小鼠肺组织胶原及羟脯氨酸(HYP)含量的影响。方法 BALB/C小鼠共32只,随机分为空白对照组、模型对照组、醋酸泼尼松组及红花水煎液组,每组各8只。除空白对照组外,其余各组均采用博莱霉素建立SSc小鼠模型,建模同时各组给予对应药物灌胃。建模后第28 d处死小鼠,取左肺制成石蜡切片进行Sirius Red染色,使用Image-pro-plus 6.0测定Sirius Red染色胶原含量,取右肺按试剂盒说明书测定HYP的含量。结果与空白对照组比较,各组小鼠肺组织胶原及HYP含量均明显升高(P<0.05);在使用醋酸泼尼松混悬液和红花水煎液治疗后,小鼠肺组织胶原及HYP含量较模型对照组显著降低(P<0.05)。结论红花水煎液能减少博莱霉素诱导的SSc小鼠肺组织胶原及HYP含量,其机制可能通过抑制肺组织胶原的沉积,从而有效防治SSc肺纤维化。Objective To study the effects of safflower decoction on collagen content and hydroxyproline(HYP) of lung in bleomycin-induced rat with systemic scleroderma.Methods 32 BALB/C rats were randomly divided into 4 groups : blank control group, model group, prednisone acetate group and safflower decoction group (8 rats in each group). Except the blank control group,the other 3 groups were used to establish rat models with systemic scleroderma by bleomycin;meanwhile, rats in the 3 groups were given intragastric administration with corresponding drugs.All the 3 groups of rats were executed,28 days after modeling.Rats' paraffin sections from the left lung were used for Sirius Red staining. Sirius Red staining collagen content was determined by Image-pro-plus 6.0 image analysis software,and the right lungs were used to determine the content of HYP according to kit instructions.Results Compared with the blank control group,the collagen content and the HYP content in the lung tissue of rats in each group significantly increased(P〈O.O5).After treatment with prednisone acetate suspension and saf-flower decoction,the collagen content and the HYP content in the lung tissue of rats significantly decreased (P〈0.05).Conclusion The Safflower edecoction can efficiently decrease the lung collagen content and the HYP content in bleomycin-induced rats with systemic scleroderma, which may be carried out by inhibiting the deposition of collagen in lung tissue ,so as to effectively prevent and treat systemic scleroderma pulmonary fibrosis.
