摘 要:目的通过测定吸烟雄鼠睾丸组织结构及细胞凋亡情况,探讨吸烟对雄性生殖系统的损伤机制。方法将清洁级SD雄性大鼠160只随机分为染毒2、4、6、8、12周的低、中、高剂量染毒组及对照组,每组10只。采用呼吸道静式染毒,每日染毒1次,染毒周期为2、4、6、8、12周。采用HE染色及末端转移酶标记技术(TUNEL法)测定大鼠睾丸组织病理学改变及细胞凋亡情况,用ELISA法检测大鼠血浆抑制素B的表达水平。结果吸烟组大鼠睾丸脏器系数从第8周起逐渐减小,与2周各剂量染毒组相比,差异有统计学意义(P<0.05)。吸烟组大鼠随着染毒时间的延长和剂量的增加,睾丸组织病理学改变逐渐明显,染毒12周后大鼠睾丸组织支持细胞呈现空泡化,管腔内观察不到游离精子。吸烟组大鼠睾丸细胞凋亡指数随染毒时间及剂量的增加而升高,染毒4周高剂量组、6周中及高剂量组、8周及12周各剂量组睾丸细胞凋亡指数均高于对照组,差异有统计学意义(P<0.05)。随着染毒时间及剂量的增加,大鼠血浆抑制素B水平逐渐降低,染毒第8周起低于对照组,差异有统计学意义(P<0.05)。结论香烟烟雾中的有害成分可致大鼠睾丸组织结构损伤及睾丸细胞凋亡率增加,并抑制血浆抑制素B分泌。Objective To explore the injury mechanism of cigarette smoking(CS) on male reproductive system by HE staining and TUNEL assay. Methods A total of 160 male SD rats were randomly divided into 15 cigarette exposure groups and one control group(without CS exposure), 10 in each, the exposure levels were 10, 20 and 30 nonfilter cigarettes perday, the exposure duration were 2, 4, 6, 8 and 12 weeks respectively. The rats were selected respectively at different time-points. The organ coefficients of testis and epididymis were calculated, testicular tissue structure and cell apoptosis were examined by HE staining and TUNEL assay. The concentrations of inhibin B in plasma were determined by enzyme-linked immuno sorbent assay(ELISA). Results The testicular organ coefficients of cigarette smoking exposure groups significantly decreased since the8 th week compared with control group(P〈0.05); The testes of rats in CS exposed groups exhibited disturbance in diverse stages of spermatogenesis and disruption of tunica propria and basement membrane, the disruption of testicular histomorphology with a time and dose dependent manner. The apoptotic positive cells were increased in TUNEL staining linked to CS exposure duration, the testicular apoptosis rate of cigarette smoking exposure groups significantly increased since the 4th week compared with control group(P 0.05). The concentrations of inhibin B in plasma of cigarette smoking exposure groups significantly decreased since the 8th week compared with control group(P〈0.05). Conclusion The harmful components in cigarette smoke may damage the tissue structure of male rats,the secretion of inhibin B and induce apoptosis of spermatogenic cells.
