DON毒素与氟对骨软骨联合损伤机制的研究

摘　　要：目的观察DON毒素和氟对TGF-β2等4种骨生长发育相关指标的影响,探讨DON毒素联合元素氟致骨软骨的损伤机制。方法 28日龄Wistar大鼠40只,按照随机化方法分为空白组、氟(F)组、DON毒素组和"DON+F"组。F组和"DON+F"组饮用150 mg/L Na F溶液,空白组和DON毒素组饮用去离子水;各组的DON毒素染毒按0.2μg/g剂量每天灌胃,空白组和F组用等量蒸馏水灌胃。喂养5周后取血,ELISA法测定各细胞因子。结果血清中TGF-β2的平均含量"DON+F"组(55.47±5.07 pg/ml)>DON组(38.88±7.99 pg/ml)>F组(38.29±5.61 pg/ml)>对照组(29.20±6.12 pg/ml);血清中BMP-2的平均含量"DON+F"组(15.24±0.66 ng/ml)>F组(14.70±1.88 ng/ml)>DON组(13.94±0.70 ng/ml)>对照组(12.44±1.07 ng/ml);血清中Hyp的平均含量"DON+F"组(7.30±1.20μg/ml)<DON组(7.38±1.65μg/ml)<F组(8.35±1.51μg/ml)<对照组(12.51±1.79μg/ml);血清中TNF-α的平均含量"DON+F"组(936.88±126.55 pg/ml)>F组(860.09±117.45 pg/ml)>DON组(706.52±73.72 pg/ml)>对照组(579.54±144.53pg/ml)。结论 DON毒素和F存在一定的交互作用,可导致染毒大鼠血清TGF-β2等细胞因子的变化,进而影响骨软骨的生长发育,导致病理损伤。Objective To observe the effects of DON toxin and fluoride on 4 bone growth and development indexes,study on the mechanism of cartilage injury. Methods Fourty fcmale were rats randomly divided into 4 groups： control,F,DON and DON ＋ F.F and DON ＋ F group, were drunk the Na F solution with 150mg/L, control and DON group were drunk with deionized solution;DON and DON ＋ F group were gavage deionized solution of DON with a dose of 0. 2 μg/g every day, control and F group were gavage with equal amount distilled water. The rats serum was taken the ELISA after 5 weeks. Results The results of ELISA showed that the concentration of TGF-β2 in the serum is DON ＋ F group（ 55. 47 ± 5. 07 pg/ml） DON group（ 38. 88 ± 7. 99 pg/ml） F group（ 38. 29 ± 5. 61 pg/ml） control group（ 29. 20 ± 6. 12 pg/ml）. The average content of BMP-2 is＂DON ＋ F＂ group（ 15. 24 ± 0. 66ng/ml） F group（ 14. 70 ± 1. 88 ng/ml） DON group（ 13. 94 ± 0. 70 ng/ml） control group（ 12. 44 ± 1. 07ng/ml）. The average content of Hyp is ＂DON ＋ F＂ group（ 7. 30 ± 1. 20μg/ml） DON group（ 7. 38 ± 1. 65μg/ml） F group（ 8. 35 ±1. 51μg/ml） control group（ 12. 51 ± 1. 79μg/ml）. The average content of TNF-α is ＂DON ＋ F＂ group（ 936. 88 ± 126. 55 pg/ml） F group（ 860. 09 ± 117. 45 pg/ml） DON group（ 706. 52 ± 73. 72 pg/ml） control group（ 579. 54 ± 144. 53 pg/ml）. Conclusions There are some interactions between the DON toxin and F. They can lead to the the changes of serum TGF-β2 and other cytokines in rats. They may affect the growth and development of osteochondral, leading to pathological damage.
