壮骨止痛方对体外培养成骨细胞Wnt/β-catenin信号通路的影响

摘　　要：目的观察壮骨止痛方对体外培养成骨细胞Wnt/β-catenin信号通路的影响。方法取新生SD大鼠颅骨进行成骨细胞原代培养,传代后随机分为4组:假手术组、模型组、对照组、中药组。培养3代后进行鉴定,加入相应含药血清干预48 h。测定各组成骨细胞Wnt3a、β-catenin、LRP5、GSK3βmRNA及蛋白的表达情况。结果与假手术组比较,模型组β-catenin、LRP5蛋白及mRNA降低(P<0.05),GSK3β蛋白及mRNA升高(P<0.05)。与模型组比较,对照组和中药组β-catenin、LRP5蛋白及mRNA表达升高(P<0.05),GSK3β蛋白表达降低(P<0.05),中药组GSK3βmRNA表达降低(P<0.05)。与对照组比较,中药组β-catenin蛋白及mRNA表达升高(P<0.05),GSK3βmRNA表达降低(P<0.05)。结论壮骨止痛方可通过上调Wnt3a、β-catenin、LRP5蛋白的表达,下调GSK3β的表达调控Wnt/β-catenin信号通路。Objective To observe the effect of Zhuanggu Zhitong Recipe（ ZGZTR） on Wnt/β-catenin signaling pathway of osteoblast culturedin vitro. Methods Osteoblasts from neonatal SD rats were cultured in primary culture then divided into 4 groups randomly,sham-operated group,model group,control group and Chinese medicine（ CM） group. Osteoblasts were identified after 3 generations,then intervened by drug containing serum for 48 h. The mRNA and protein expression of Wnt3 a,β-catenin,LRP5,GSK3β were detected. Results Compared with sham-operated group,the protein and mRNA expression of β-catenin,LRP5 decreased（P 0. 05）,GSK3β mRNA and protein increased in model group（P 0. 05）. Compared with the model group,the mRNA and protein expression of β-catenin,LRP5 increased（P 0. 05）,the protein expression of GSK3β decreased in control group and CM group（P 0. 05）; the mRNA expression of GSK3β decreased in CM group（P 0. 05）. Compared with the control group,the mRNA and protein expression of β-catenin increased（P 0. 05）,and GSK3β mRNA expression decreased（P 0. 05）in CM group. Conclusion ZGZTR can regulate the Wnt/β-catenin signaling pathway by raising the expressions of protein Wnt3 a,β-catenin,LRP5 and reducing the expression of GSK3β.
